| BackgroundEsophageal cancer is one of the most common digestive carcinoma in our country;the symptom of esophageal cancer in early stage is not so special that the disease is usually in advanced stage when it is diagnosed clearly.Therefore it has a higher mortality,and located in the fourth of all kinds of malignant tumors,and the depth of invasion and metastasis are the main reason of tumor-related deaths.At present,the treatment of esophageal cancer includes surgical procedure,radiotherapy and chemotherapy.But,with the better understanding of cancer pathogenesis in molecular level,aim at the specific molecular target of new molecular targets has become a new direction for the treatment of esophageal cancer.Astrocyte elevated gene-1 is the newly discovered oncogene.The study found that AEG-1 has high expression in a variety of tumor s includes breast cancer,glioma,prostate cancer,stomach cancer,and it’s closely related to tumor progression,invasion and metastasis and poor prognosis.E-cadherin is a adhesion molecules that expressions in the cytomembrane of normal epithelial cells,and it mediated adhesion between epithelial cells.In varieties of tumors,the expression of E-cadherin is significantly reduced or even missing,and it’s closely related to tumor invasion and metastasis and poor prognosis.However,the correlation of expressions of AEG-1 and E-cadherin in esophageal cancer has not been reported.This research proposed to detect the expression level of AEG-1 and E-cadherin in esophageal carcinoma tissues and corresponding adjacent carcinoma tissues by immunohistochemistry and Western blot,analyze its relationship with clinical pathological features,to explore the correlation of both expressed in esophageal cancer and its significance.ObjectiveTo explore the expression correlation of astrocyte elevated gene-1(AEG-1)and E-cadherin(E-ca)protein in esophageal carcinoma,and analyze its relationship with clinical pathological features.To explore the role of AEG-1 and E-ca in the invasion and metastasis of esophageal carcinoma.MethodsImmunohistochemical and Western Blot test methods were used to detect the expression of AEG-1 and E-cadherin protein in 58 cases of esophageal carcinoma tissues and its corresponding adjacent tissues.,and collect the complete pathological data.Then we analysed statistically the correlation among them with clinic-pathological differentiation status,TNM stage,tumor size and so on.The data were analyzed by using SPSS 19.0 statistical software,count data using χ 2 test,Correlation using Spearman correlation analysis,α=0.05 as tests,with P<0.05 for the difference was statistical significant.Results1.In 58 cases of esophageal carcinoma tissues,the positive expression rate of AEG-1was 82.7%;The positive expression rate of AEG-1 was 51.7% in the corresponding adjacent tissues,the positive expression rate of AEG-1 in esophageal carcinoma tissues was obviously higher than that in the corresponding adjacent tissues(P<0.05),and the expression positive rate of AEG-1 in esophageal carcinoma tissues was related to differentiated degree,depth of invasion,clinical stage,and lymph node metastasis(P<0.05);but no relation with sex,age,tumor site,and pathologic types(P >0.05).2.in 58 cases of esophageal carcinoma tissues,the positive expression rate of E-cadherin protein was 31.0%,the positive expression rate of E-cadherin was 94.8%;the positive expression rate of E-cadherin in esophageal carcinoma tissues was obviously less than that in the corresponding adjacent tissues(P <0.05);it’s related to differentiated degree,depth of invasion,clinical stage,and lymph node metastasis(P <0.05);but no relation withsex,age,tumor site,and pathologic types(P >0.05).3.the expression of AEG-1 and E-cadherin protein was significantly negative correlation(r=0.483,P <0.05).Conclusion1.The expression of AEG-1 and E-cadherin participate in the process of esophageal cancer invasion and metastasis,Joint detection the expressing of AEG-1 and E-ca is the important factor of patients with esophageal cancer metastasis potential and prognosis.2.AEG-1 may be mediated E-cadherin less-expression in esophageal carcinoma. |
