Objective:To observe the effect of acupuncture combined hypothermia therapy in Cerebral ischemia reperfusion injury Rats about neural function defect scale, cerebral infarction area ratio, p-Raf1, p-MEK2, p-ERK1/2 on the MAPK/ERK pathway and cell apoptosis, explore the mechanism of acupuncture combined low hypothermia therapy for ischemic stroke.Methods:Reference Zea Longa line tied to copy the MCA ischemia model(MCAO) and modified, 90 SD rats were randomly divided into blank, control, model, acupuncture(acupuncture point take the Dazhui(GV14), Baihui(GV20), Renzhong(GV26)), mild hypothermia, the acupuncture combined low hypothermia, after 72 h treatment, observe the nerve function defect grade and TTC staining detection infarction area ratio, TUNEL method to detect apoptosis cells, western blot method to detect the expression level of p-Raf1, p-MEK2, p-ERK1/2 in the rat hippocampus ischemia side.Results: 1.The comparision of neurological deficit score in each group rats: Before treatment, compared with the blank group and control group, nerve function score of the rest of the group rats were significantly higher(P<0.01); After treatment, compared with model group, nerve function score of acupuncture group, mild hypothermia group and acupuncture combined hypothermia group was lower(P<0.01); Compared with acupuncture group, nerve function score of acupuncture combined hypothermia group was lower(P<0.01); Compared to the rest between each treatment group, there was no statistically significant difference(P>0.05). 2.The comparison of each cerebral infarction area ratio: compared with the blank and control group, cerebral infarction area ratio of the rest of the group rats were significantly higher(P<0.01); Compared with model group, cerebral infarction area ratio of acupuncture group, mild hypothermia group and acupuncture combined hypothermia group was lower(P<0.05 or P<0.01); Between each treatment group compared with no statistically significant difference(P>0.05). 3.The comparison of each lateral ischemic brain tissue cell apoptosis:compared with the blank group and control group, apoptotic cells of the rest of the group rats were significantly higher(P<0.05); Compared with model group, apoptotic cells of acupuncture group, mild hypothermia group and acupuncture combined hypothermia group was lower(P<0.01); Between each treatment group compared with no statistically significant difference(P>0.05). 4.Groups of ischemia bilateral hippocampal tissues phosphorylation Raf1, MEK2, ERK1/2 expression level of comparison: Compared with the blank group and control group, model group, acupuncture group, mild hypothermia group and acupuncture combined hypothermia group could increase p-Raf1, p-MEK2, p-ERK1/2 expression in the rat hippocampus(P<0.05 or P<0.01); Compared with model group, p-Raf1, pMEK2, p-ERK1/2 expression of the treatment group in the rat hippocampus were lower (P<0.01); Compared with acupuncture group, p-Raf1, p-MEK2, p-ERK1/2 expression of the low temperature in the rat hippocampus were no decrease(P>0.05), and p-Raf1, p-MEK2, p-ERK1/2 expression of acupuncture combined hypothermia group in the rat hippocampus reduced(P<0.01 or P<0.05); Compared with mild hypothermia group, p-Raf1, p-MEK2, p-ERK1/2 expression of acupuncture combined hypothermia group in the rat hippocampus significantly decreased(P<0.01).Conclusion : 1.Acupuncture, mild hypothermia and acupuncture combined hypothermia group can improve nerve function defect, reduce the cerebral infarction area and cell apoptosis, protection effect to the brain tissue. 2.Its protective mechanism of brain tissue may be through the MAPK/ERK pathway, reduce the phosphorylation Raf-1, MEK-2, ERK1 /2 protein expression levels. 3.In reducing the neural function defect scale and reduce p-Raf1, p-MEK2, p-ERK1/2, acupuncture combined hypothermia group is better than the acupuncture group or the low temperature group, may be because the acupuncture therapy and mild hypothermia therapy superposition or synergy effect.
