Objective:The effects of acupuncture at Dazhui,Baihui and Renzhong acupoints on neurological deficit score,cerebral infarct size ratio and phosphorylation of ES,VEGF and ERK1/2 protein in rats with cerebral ischemia-reperfusion injury were investigated.Part of the mechanism of brain protection after cerebral ischemia-reperfusion injury.Methods:There were 100 SD males in the rats.After serialization,40 randomly selected according to the random number table method,and then randomly divided into normal group and sham operation group,20rats/group,and the remaining 60 rats were referred to Zea Longa modified line plug method.The middle cerebral artery occlusion(MCAO)was prepared and randomly divided into model group,drug group and acupuncture point group,20 rats/group.After the vital signs of the rats to be modeled were stable,the normal group did not do any treatment.The rats in the sham operation group and the model group were only bundled without acupuncture,bundled for 30 min,12 h/time;the rats in the drug group were bundled at 3 mg/kg(diluted to 1mL)dose of edaravone was intraperitoneally injected,bundled for 30 min,12h/time;rats in the acupoint group were acupunctured at Dazhui,Baihui,and Zhongzhong three points,each point was rotated for 60 s,and the needle was placed once every 15 minutes.30 min,12 h/time.The rats were scored for neurological deficits before and after the first treatment.The rats were sacrificed after 6treatments.The whole brain slices were taken and the ratio of cerebral infarction was observed by TTC staining.The hippocampus was isolated from the ischemic side.The expression levels of p-ES,p-VEGF and p-ERK1/2 were detected by blot.Results:Before treatment,the neurological deficit scores of the model group,the drug group and the acupuncture group were significantly higher than those of the normal group and the sham operation group,and there was a statistically significant difference(P<0.01),indicating successful modeling;comparison between the three groups There was no statisticaldifference.After treatment,the neurological deficit scores of the model group,the drug group and the acupuncture group increased compared with the normal group and the sham operation group,the difference was statistically significant(P<0.01);compared with the model group,the drug group and the needle point The scores of neurological deficits decreased,and the difference was statistically significant(P<0.01).There was no significant difference between the acupuncture group and the drug group.There was no difference in the neurological deficit scores between the normal group and the sham-operated group before treatment;there was no significant difference in the neurological deficit scores in the model group,and the neurological deficit scores in the drug group and the acupuncture group decreased.Statistical difference(P<0.01).At the same time,compared with the normal group and the sham operation group,the cerebral infarct size ratio of the model group,the drug group and the acupuncture group increased,and the difference was statistically significant(P<0.01).Compared with the model group,the ratio of cerebral infarction area in the drug group and the acupuncture group was decreased,and there was a statistically significant difference(P<0.01).There was no significant difference in the area of cerebral infarction between the acupuncture group and the drug group.Finally,compared with the normal group and the sham operation group,the expressions of p-ES,p-VEGF and p-ERK1/2 in the hippocampus of the model group,the drug group and the acupuncture group were all increased,and the difference was statistically significant(P<0.01 or P<0.05).Compared with the model group,the expression of p-ES was decreased in the drug group and acupoint group,and the expressions of p-VEGF and p-ERK1/2 were all increased,the difference was statistically significant(P<0.01 or P<0.05).There was no significant difference in the expression of p-ES,p-VEGF and p-ERK1/2 between the acupuncture group and the drug group.Conclusion:Acupuncture at Dazhui,Baihui and Renzhong acupoints can alleviate the symptoms of neurological deficits in rats after cerebral ischemia-reperfusion injury,and reduce the area of cerebral infarction,so as to achieve therapeutic purposes.Its mechanism may be related todown-regulating the expression of p-ES in the ischemic region,reducing its inhibitory effect on VEGF-mediated angiogenesis,up-regulating the expression of p-VEGF and p-ERK1/2,activating VEGF signaling pathway,and bidirectionally regulating the positive and negative factors of angiogenesis.To guide angiogenesis related.
