Objective: The purpose of this study was to investigate the clinical effect about expressions of STC2,MMP-9 and PCNA in esophageal squamous cell carcinoma,explore the relation with clinical pathology and prognosis,and provide molecular and reference basis for clinical staging of esophageal carcinoma.Methods: Experimental materials were selected from the Department of Thoracic Surgery in our hospital.There were 54 postoperative samples in whom had 36 male cases and 18 female cases,aging from 26 to 73,without neoadjuvant therapy.There were also 20 adjacent carcinoma tissues(beside carcinoma 1~3cm)and 16 corresponding esophageal normal mucous membrance.Use immunohistochemical method to test the expressions of STC2,MMP-9 and PCNA in 54 cases of esophageal carcinoma,20 cases of adjacent carcinoma tissues and 16 cases of normal esophageal mucosa.Find the relationship between the expressions and clinical pathological parameters.Results: The expressions of STC2,MMP-9 and PCNA in normal esophageal mucosa,adjacent carcinoma tissues and esophageal carcinoma tissues were 31.3%,45.0%,72.2% and 6.3%,40.0%,79.6% and 18.8%,25.0%,70.4%.The expressions of the three indicators were gradually enhanced with statistically significant(P<0.05).Three indicators' expressions in esophageal cancer tissue were significantly higher than its expression of tissue adjacent to carcinoma and normal esophageal mucosa(P < 0.05),but the expressions of adjacent carcinoma tissues and normal esophageal mucosa showed no significant difference(P > 0.05).In esophageal carcinoma tissue,the expressions of STC2,MMP-9 and PCNA with lymphatic invasion,lymph node metastasis,poorly differentiated and pathological stage III ~ IV were significantly higher than that opposite ones,and the difference was statistically significant(P<0.05).In STC2 positive group,positive expressions of MMP-9 and PCNA were significantly higher than that of STC2 negative group,and the difference was statistically significant(P<0.05).Besides,expressions of PCNA and MMP-9 were positively correlation with STC2 expression.Multivariate analysis showed that histological grading(OR=2.582,P<0.05)and STC2(OR=0.354,P<0.05)were independent factors affecting the prognosis of esophageal carcinoma.While age,gender,lymphatic invasion,pathological staging,MMP-9 and PCNA expressions did not affect the prognosis of independent factors.The survival of patients with STC2,MMP-9 and PCNA positive expression were significantly shorter than that of the patients with negative expression,and the difference has statistical significance(?~2=5.112,4.111,4.618,P < 0.05).Esophageal carcinoma patients' expressions of STC2,MMP-9,PCNA with two indicators positive expression had significantly shorter survival than the other patients,and the difference was statistically significant(?~2=7.682,9.531,11.097,P < 0.01).Conclusions: The expressions of STC2,MMP-9 and PCNA in esophageal carcinoma are significantly higher than that of adjacent carcinoma tissues and normal mucous membrance tissues,and correlated with lymphatic invasion,lymph node metastasis,high histological grade and high pathological stage.MMP-9 and PCNA expressions are positively correlated with STC2 expression.STC2 is an independent prognostic factor for esophageal carcinoma.The expressions of STC2,MMP-9 and PCNA are higher in the patients with poor prognosis.
