Study On The Chemical Component And Quantity Standard For The Herbs Of Pilea Sinofasciata C.J.Chen

Objectives: Study on the chemical component of ethyl acetate extract and n-butanol extract for the herbs of Pilea sinofasciata C.J.Chen.To establish a TLC method for identifying of Pilea sinofasciata C.J.Chen in different areas.According to the testing methods in the 2015 edition of Chinese Pharmacopoeia,study on quality standard for Pilea sinofasciata C.J.Chen and establish a method for assessment of Pilea sinofasciata C.J.Chen.The method can be used scientifically as a quality control for Pilea sinofasciata C.J.Chen.Methods: The alcoholic extracts obtained from Pilea sinofasciata C.J.Chen's dried herbs were divided into three portions by systematic solution:petroleum ether extract,ethyl acetate extract and n-butanol(saturated with water)extract;thin layer chromatography,Silica gel column chromatography,sephadex column chromatography,high performance liquid chromatography preparation,recrystallization,Medium pressure liquid chromatography and other modern techniques and methods were used to isolate and purify the ethyl acetate extract and n-butanol extract.Pury compounds were identified by physical and chemical,IR ? MS ? 13C-NMR ? 1H-NMR,et al.For Pilea sinofasciata C.J.Chen,done the mensuration of moisture,total ash,acid-insoluble and content testing to ethanol extracts in the 2015 edition of Chinese Pharmacopoeia.With Hernandulcin as the references substances,the TLC conditions,such as extraction solvents,extraction methods,chromogenic agent,developing solvent and temperature/humidity were investigated.Combined the stipulationg under Pilea sinofasciataC.J.Chen in the 2015 edition of Chinese Pharmacopoeia,the initial quality standard draft were established.Results: The ethyl acetate extract and n-butanol extract obtained from the herbs of Pilea sinofasciata C.J.Chen were separated.Twenty compounds were separated and sixteen were identified as ethyl linoleate(1),palmitic acid ethyl ester(2),?-amyrin(3),palmitic acid(4),eicosanoid(5),lignoceric acid(6),daucosterol(7),heptacosanic acid(8),adenosine(9),Indole-4-carboxylicacid(10),protocatechuate(11),gallic acid(12),betulinic acid(13),stearic acid(14),oleanolic acid(15),potassium nitrate(16).Through the study of Pilea sinofasciata C.J in different areas,The moisture content does not exceed 12%,Total ash is not allowed to be22%,Acid insoluble ash for not more than 3.5%and the extract content is not less than 24%.A good linear relationship between the medicinal herbs,RSD values were in the normal range.In accordance with the General rules 0502 of the fourth part Chinese Pharmacopoeia(2015 edition),to study the thin layer chromatographic with Hernandulcin as the references substances of Pilea sinofasciata C.J.Chen in different locations.The best thin layer chromatographic condition is the ethanol ultrasound extraction under the interesting for 40 min at 20? with the humidity of 77%,it was performed on a TLC plate with Petroleumether-Trichloromethane-Acetone(7:1:1).To draw 5 ?L contrast medicinal materials and standard products point on the same silica gel G thin layer plate,take out after the expansion and dry,and then sprayed with 10%sulphuricacid and ethanol solutionas,dried at 105 Celsius degrees,the spots were clear and the resolution was good.The results showed that medicinal materials containing the Novel Sweetener Hernandulcin for Pilea sinofasciata C.J.Chen,the Rf value was about 0.35.Conclusion(s): Twenty compounds were separated and sixteencompounds were identified from the ethyl acetate extract and n-butanol extract of Pilea sinofasciata C.J.Chen.All of the compounds were isolated from this plant for the first time.Using the thin layer chromatographic with Hernandulcin as the references substances of Pilea sinofasciata C.J.Chen in different locations,experiments show that this method is simple,high repeatability and accurate result.The results of thin layer chromatography and inspection results provided the evaluation index for the quality control of Pilea sinofasciata C.J.Chen.Combined the stipulationg under Pilea sinofasciata C.J.Chen in the 2015 edition of Chinese Pharmacopoeia,the initial quality standard draft were established,The standard is scientific,feasible,practical and so on,to provide reference for quality control of Pilea sinofasciata C.J.Chen.
