In order to study the quality standards of Sichuan rhubarb,and provide the planting theoretical for the local farmers,this paper established a simple and effective rhubarb fingerprint,five kinds of active substances of four rhubarb is analyzed,and identified genetic relationship of four types of rhubarb.12 high performance liquid chromatography(HPLC)figures of three batches of four kind of rhubarb(R.palmatum,R.palmatum var.tanguticum,R.officinale,R.rhaponticum)were used to establish rhubarb's fingerprint.We choused a characteristic peak to evaluate the similarity of fingerprint,the RSD of retention time of common peak were between 0.05% and 4.63% and the RSD of peak area were between 1.74% and 151.19%.This indicated that the 12 sample's composition was similarly,but its content of each component was different.The evaluation results showed that the similarities were: 0.97,0.97,0.96,0.94,0.9,0.97,0.93,0.92,0.9,0.97,0.91,0.97,indicated that there was good similarity of fingerprint,it can be used to qualitative analysis and the identification of rhubarb.To evaluate the quality of four kind rhubarbs,we use established fingerprint of rhubarb to identify the four commercially rhubarbs and high performance liquid chromatography(HPLC)to measure the contents of five kinds of active substances(rhein;Chrysophanol;emodin;Aloe-emodin;Physcion)in four kind rhubarbs.The results showed that the HPLC peak of four commercial rhubarbs were accordance with the fingerprint chromatogram peak;the content of emodin was highest in R.palmatum,the content of chrysophanol was the highest in R.rhaponticum,the content of rhein was the highest in R.palmatum var.tanguticum,the content of Physcion was the highest in R.rhaponticum,and the content of Aloe-emodin was the highest in R.officinale.Because of the HPLC of R.rhaponticum and R.palmatum was similar,R.rhaponticum did not enter the pharmacopoeia,but it is widely used in the market.To explore that,we identified the genetic relationship of the four kinds of rhubarb.The results showed that the genetic relationship of R.rhaponticum and R.palmatum were closest,which was consistent with the fingerprint results.Which provided the theoretical basis for the phenomenon of R.rhaponticum did not enter the pharmacopoeia but it is widely used in the market.It also provided data support for R.rhaponticum to enter pharmacopoeia.
