Background:Phlomis umbrosa Turcz.,is one of the plants of the genus Phlomis(Lamiaceae).And there are about 100 species of Phlomis in the world which distributed mainly in the Mediterranean region and eastwards to the western Himalayas.Fourty-one species can be found in most region of China,especially in southwest of China.Its herb has been used in Chinese traditional medicine,their taste flavour WenXin,can wind,and it is a kind of commonly used medicinal plants.So far,phytochemical studies of P.umbrosa were reported previously,but no research about chemical constituents of Lingkong Mountain in Shanxi Province has been reported.The genus Isodon is a perennial herbs of Labiatae as well,can be found in most every region of China.The leaves of Isodon japonica have been used as an antibacterial,anti-inflammatory,stomachic,and anthelmintic agent in Chinese traditional medicine.At present,more than 80 species of the genus Isodon have been researched and over 500 diterpenoids have been isolated from this genus.However,Isodon japonica which is from Lingbao in Henan Province has not been reported.Objective:To study the constituents and biological activities of Phlomis umbrosa Trucz.and Isodon japonica.Method:Using modern separation methods to extract and separate the chemical components such as silica gel column chromatography(CC),preparative thin layer chromatography(PTLC)and high performance liquid chromatography(HPLC).And using modern spectra technology such as IR,UV,1H-NMR,13C-NMR,DEPT,1H-1H COSY,HMQC,HMBC,NOESY,EI-MS and other physical and chemical methods to analyse and appraise the monomer compounds,finally to confirm the structure of the compounds.The in vitro anti-tumour activities of the compounds from Isodon japonica were studied by RSB assay.Result:Among the 25 compounds isolated from the air-dried and powdered leaves of Phlomis umbrosa Turcz.,two new dammarane-type glycosides,two new iridoid compounds,seven flavonoids and its glycosides compounds,eight iridoid compounds,and six other compounds.All these compounds were as follows:3?,23S*,25-trihydroxy-20S*,24S*-epoxydammarane 3-O-[?-D-glucopyranosyl(2?1)-?-L-rhamnopyranosyl]-23-O-?-D-glucopyranoside(1)?3?,235*,24R*-trihydroxy-20S*,25-epoxydammarane 3-0-[?-D-glucopyranosyl(2?1)-a-L-rhamnopyranosyl]-23-O-?-D-glucopyranoside(2)?(1S*,3R*,4S*,5S*,6R*,7R*,8S*,9S*)-3,8-epoxy-1-hydroxy-4-methoxycarbonyl-10-methyl-6,7-a cetonide-cis-2-oxabicyclo[4.3.0]nonane(3)?(1R*,3R*,4S*,5S*,6R*,7R*,8S*,9S*)-3,8-epoxy-1-hydroxy-4-methoxycarbonyl-10-methyl-6,7-acetonide-cis-2-oxabicyclo[4.3.0]nonane(4),lamiophlomiol A(5),lamiophlomiol B(6),8-acetylshanzhigenin methyl ester(7),8-acetyl-1-epi-shanzhigenin methyl ester(8),shanzhigenin methyl ester(9),1-epishanzhigenin methyl ester(10),· 1?-hydroxy-dihydrocornin aglycone(11),1?-hydroxy-di-hydro-comin aglycone(12),loliolide(13),apigenin(14),luteolin(15),apigenin-7-O-?-glucoside(16),luteolin-7-O-?-glucoside(17),apigenin-7-O-?-D-glucuronide methyl ester(18),aigenin-7-O-(6"-O-p-coumaroyl)-?-D-glucopyranoside(19),apigenin-7-O-(3"-p-courmaryl)-glucoside(20),Succinic acid(21),Vanillic acid(22),3,4-dimethoxy-benzoic acid(23),Salicylic acid(24),methyl ?-D-glucoside(25).Fifteen compounds identified from Isodon japonica are Oridion(1),cetonide of maoyecrystal F(2),Effusanin A(3),Lasiodonin(4),Enmenol(5),Enmein(6),Nodosin(7),Eriocaside A(8),Oleanolic acid(9),2?,3?-dihydroxyolean-12-en-28-oic acid(10),3?-Hydroxy-5a-cholesta-6,8(14)-diene(11),Stigmasterol(12),?-Sitosterol(13),?-daucosterol(14),Succinic acid(15).Effusanin A(3),Enmein(6)and Nodosin(7)show significant inhibition to proliferation of tumour cell,HL-60,SMMC-7721,HeLa.Conclusion:Among the 25 compounds were isolated from Phlomis umbrosa,4 new compounds,5 compounds isolated from this genus for the first time and 2 compounds isolated from this plant for the first time;Effusanin A(3),Enmein(6)and Nodosin(7)which were isolated from Isodon japonica exhibited potential anti-tumour activities.
