Objective:To study the effects of three peptides KM-5?13?14 on platelet aggregation and activation functions stimulated by collagen or arachidonic acid(AA) respectively.Methods:Turbidiemetric platelet aggregometry and flow cytometry were used to test respectively the effects of three peptides KM-5?13?14 on platelet aggregation rates and the CD62p expression of platelets stimulated by collagen or AA.Results:1?Turbidiemetric platelet aggregometry:?Compared with the collagen group, the aggregation rates of groups KM-5 (5nM/10nM/20nM)+collagen were higher, at the concentration 5nM of KM-5, the promoting effect was more apparent, platelet aggregation rate increased 18%, but there had no statistically difference (40.67±5.03% vs.58.00±11.53%, n=3,p=0.204 >0.05), KM-5 had no obvious effect on platelet aggregation stimulated by AA. (p=0.912>0.05);?Compared with the collagen group, the aggregation rates of groups KM-13 (5nM/10nM/20nM)+collagen were higher, at the concentration 5nM of KM-13, the promoting effect was more apparent, platelet aggregation rate increased 8%, but there had no statistically difference (33.67±8.50% vs.41.67±12.06%, n=3, P=0.666> 0.05), KM-13 had no obvious effect on platelet aggregation stimulated by AA (p=0.978>0.05).?Compared with the AA group, the aggregation rates of groups KM-14 (5nM/10nM/20nM)+AA were lower, the effects showed dose-dependent, at the concentration 20nM of KM-14, the inhibiting effect was more apparent, platelet aggregation rate reduced 10%, there had no statistically difference (27.67±10.02% vs. 17.00±6.08%, n=3,p=0.522>0.05), KM-14 had no obvious effect on platelet aggregation stimulated by collagen. (p=0.901>0.05)2?Flow cytometry:? KM-5 (5nM/10nM/20nM) had no obvious effect on the expression of CD62p stimulated by AA/collagen(pAA=0.108>0.05;pcollagen=0.381>0.05).? Compared with the collagen group, groups KM-13 (5nM/10nM/20nM) +collagen could significantly increase the CD62p expression, at the concentration 20nM of it, the expression rate increased 26%, but there had no statistically difference(28.70±7.92% vs.54.00±17.68%, n=2, p=0.212>0.05), groups KM-13 (5nM/10nM/20nM) + AA had no obvious effect on the expression of CD62p (p=0.953>0.05).? KM-14 (5nM/10nM/20nM) had no obvious effect on the expression of CD62p stimulated by AA/collagen (pAA=0.727> 0.05;pcollagen=0.623> 0.05)Conclusions:KM-14 had a inhibition trend of platelet aggregation stimulated by AA; KM-5 had a promotion trend of platelet aggregation stimulated by collagen; KM- 13 had a promotion trend of platelet aggregation and CD62p expression stimulated by collagen.
