Objective:By experimentally studying on the tumor suppression effects and anti-angiogenesis of Yanghe Decoction in S180 tumor-bearing mice,observing the influence of Yanghe Decoction on the weight,tumor weight,thymus index,thymus index,VEGF,HIF-1? protein expression,we have explored the significant important role and action target of Yanghe Decoction in the treatment of malignant tumor and discussed the action mechanism of Yanghe Decoction in treating S180 tumor-bearing mice.Methods:Firstly,the blank control group was randomly selected.Secondly,mice after inoculation were divided into five groups: tumor-bearing control group,group of Yanghe Decoction in low-dosage,group of Yanghe Decoction in medium-dosage,group of Yanghe Decoction in high-dosage and cyclophosphamide?CTX?group.The blank control group was injected with 0.9% of sodium chloride injection,equivalent to that of other groups,so did the tumor-bearing control group.Three groups of Yanghe Decoction were respectively given a gavage with certain dose of Yanghe Decoction.CTX group was given 25mg/kg of intraperitoneal injection at the first five days,0.2m1 for each,0.9% of sodium chloride injection after five days.Weight them once every three days and calculate and give the corresponding dosage according to their weights.Observe animals' general condition after giving medicines.Give medicines once daily for fifteen days.Break the neck of mice to death the next day after stopping medicine,record the body weight changes of mice before and after medicine,record tumor weight,thymus and spleen weight after medicine,collect data and calculate the anti-tumor rate,thymus and spleen index.Microscopically observe the pathologic change of tumor tissue after tissue section staining and detect the VEGF,HIF-1? protein expression by the method of immunohistochemistry.Results:1.Mice in blank control group showed vivacious movement,good appetite,agile reaction and weight gain.Mice in tumor-bearing control group showed apathetic in spirit,fast growing of tumor tissue under axilla,poor appetite,messy and reluster hair and obvious piloerection.Mice in groups of Yanghe Decoction in different dosage showed lively condition,slow growing of tumor tissue,glossy hair,less piloerection and good appetite.Mice in CTX group stayed in sleepiness,slowest growing of tumor tissue,but worse than other groups in integrated state.2.When compared with that of the blank control group after the treatment,the spleen indexes of Yanghe Decoction group in low-dosage and in medium-dosage were significantly increased,showing statistical significance?P<0.01?,and only the thymus index of Yanghe Decoction group in medium-dosage was significantly increased,showing statistical significance?P<0.01?.When compared with that of the tumor-bearing control group,the spleen indexes of all the groups were obviously increased?P<0.01?,and the thymus indexes of Yanghe Decoction group in medium-dosage and in high dosage were significantly increased,showing statistical significance?P<0.01?.When compared with that of CTX group,the spleen indexes and thymus indexes of S180 tumor-bearing group in three Yanghe Decoction groups were significantly increased and showed significant differences between groups?P < 0.05?.3.Microscopically: tumor cells of S180 tumor-bearing group arranged intensively,rich in blood vessels within the tumor tissue.There were a large number of new vessels,especially in Yanghe Decoction in medium-dosage and high-dosage.There were flake necrosis areas in each treatment group and less tumor tissue and ambient new vessels.4.VEGF,HIF-1? protein expression in tumor tissue of S180 tumor-bearing mice of each treatment group were significantly decreased and showed significant difference when compared with that of tumor-bearing group?P<0.01?.However,best effects appeared in CTX group.There was no difference between Yangehe Decoction group in medium-dosage and Yangehe Decoction group in high-dosage?P>0.05?.Conclusion:Yanghe Decoction can significantly improve the survival quality of the S180 tumor-bearing mice and inhibit the growth of S180 sarcoma in mice tumor tissue.The tumor suppression effects of Yanghe Decoction are insufficient when compared with that of CTX group.Yanghe Decoction can reduce the immune organ damage of mice and improve the mice immune function.Yanghe Decoction can down-regulate the VEGF,HIF-1?protein expression of tumor tissue in S180 tumor-bearing mice,decrease the blood and oxygen supply for tumor tissue which can inhibit tumor angiogenesis.
