Objectives Observing acupuncture and moxibustion affects on the content of stromal cell derived factor-1 (SDF-1) and lymphocyte associated antigen-1(LFA-1) in mouse marrow organization of myelosuppression model caused by Cyclophosphamide (CTX). In molecular biology level reveal the mechanism that acupuncture and moxibustion repair and protection marrow hematopoietic microenvironment after chemotherapy, so that interpret the biology mechanism that acupuncture and moxibustion maintain hematopoiesis and promote WBC count in Peripheral blood. So that provides the basis of the molecular biology laboratory for the clinical application of acupuncture and moxibustion to protect hematopoietic function in patients with chemotherapy and increase WBC in Peripheral blood.Methods The 80 SPF male KM mice were randomly divided into 3 days and 5 days groups by body weight according to the different number of treatment days. Then each group in compliance with the weight were randomly divided into blank group, model group, acupuncture group and moxibustion group, every subgroup of 10 mice. The model of myelosuppression was made with Cyclophosphamide. After the model is successfully established,in the acupuncture group and the moxibustion group,acupoints "Dazhui"(GV 14), "Geshu" (BL 17), "Shenshu" (BL 23) and "Zusanli" (ST 36) were respectively used for treatment with acupuncture and moxibustion at the same time of day.According to the different groups, treatment 3 or 5consecutive days respectively.while, in the blank group and the model group, there were no treatment carried out except for catching and fixing. Sample tail venous blood of each group mice everyday at the same time by the same person, and count WBC in Peripheral blood by microscopy. Enzyme-linked immunosorb-ent assay to detect the SDF-1 and LFA-1 content in mice marrow cells.Then using SPSS 20.0 for Windows statistical software analyzes data.Results 1 Respectively compare with the blank group, the number of white blood cells was significantly decreased(P< 0.05) in 3 days and 5 days groups which group were after cyclophosphamide (CTX) chemotherapy, showed that the two groups were successful modeling.After 3 days treatment,, acupuncture group, moxibustion group were lower than that of blank group (P< 0.05). Treatment for 5 days, white blood cell count of the model group is lower than that of the blank group (P< 0.05), while white blood cell count of acupuncture group, moxibustion group were higher than that of model group and blank group (P< 0.05).Among Model group,white blood cell was the highest before modeling, and then decreased, at the day of treatment for 3 days fell to the lowest, and then rise.The white blood cells of Acupuncture group, moxibustion group was the highest before modeling, and then decreased, after treatment for 2 days fell to the lowest, then rise.2 After modeling, the SDF-1 content in accept CTX chemotherapy group was significantly lower than that of blank group(P< 0.05), prompted CTX inhibits the expression of marrow chemokine SDF-1. The SDF-1 content variation was very similar between treatment for 3 days and 5 days.Compared with the blank group, SDF-1 content of model group, acupuncture group, moxibustion group decreased (P< 0.05). However, the SDF-1 content of acupuncture group and moxibustion group was significantly higher than that of the model group (P< 0.05), compare with acupuncture group, the SDF-1 content of moxibustion group was slightly higher but there was no statistically significant difference(P> 0.05).3 After modeling, the LFA-1 content in accept CTX chemotherapy group was significantly higher than that of blank group(P< 0.05), prompted CTX promote the expression of marrow adhesion molecule LFA-1. The LFA-1 content variation was substantially the same between treatment for 3 days and 5 days. Compared with the blank group, LFA-1 content of model group, acupuncture group, moxibustion group increased (P< 0.05). However, the LFA-1 content of acupuncture group and moxibustion group was significantly lower than that of the model group (P< 0.05). The differences between treatment for 3 days and 5 days were that:The LFA-1 content of moxibustion group was less than that of acupuncture group after treatment for 3 days but there was no statistically significant difference(P> 0.05), The LFA-1 content of moxibustion group was less than that of acupuncture group after treatment for 5 days and the difference was statistically significant (P< 0.05).4 The influence of different treatment days on the content of SDF-1 and LFA-1. The difference of SDF-1 and LFA-1 content in mice bone marrow between acupuncture for 3 days and for 5 days was not statistically significant (P> 0.05). The SDF-1 content was also no statistically significant difference in mice bone marrow between moxibustion for 3 days and for 5 days (P> 0.05).But the LFA-1 content in mice bone marrow of moxibustion 5 days is less than that of acupuncture 3 days (P< 0.05).Conclusions 1 Acupuncture treatment can confront bone marrow suppression caused by CTX chemotherapy, enhance the bone marrow hematopoietic function, improve the peripheral white blood cell count.2 Acupuncture and moxibustion can enhance hematopoietic stem/progenitor cells chemotaxis by increase expression of chemokine SDF-1 in mouse bone marrow after CTX chemotherapy. And adjust the migration of hematopoietic stem cells in bone marrow, promote early blood cells generated and proliferation, have the repair of bone marrow hematopoietic microenvironment injury after chemotherapy.3 Acupuncture and moxibustion can decrease expression of adhesion factor LFA-1 in bone marrow of miceafter CTX chemotherapy, reduce hematopoietic stem cells and bone marrow stromal cell strong adhesion, promote hematopoietic progenitor cell proliferation and differentiation. So that protect bone marrow hematopoietic microenvironment, maintain bone marrow hematopoietic function after chemotherapy.4 Compared to acupuncture and moxibustion, moxibustion is more conducive to reducing the hematopoietic progenitor cells and matrix adhesion by decreasing the content of marrow adhesion factor LFA-1 in CTX chemotherapy mouse, antagonistic inhibitory regulating factors of hematopoietic progenitor cell damage.
