Screening Of Core Herbal Combinations In Anti-alzheimer Prescriptions By Data Mining Methods

Senile dementia also is known as Alzheimer's disease. Once considered the most effective anti – Alzheimer drug tacrine was replaced because of its high side effects. Those drugs such as donepezil, galantamine and other medicines were not ideal anti – Alzheimer drugs yet, although they were improved in terms of the safety and efficacy. None of natural pure compound drugs such as Huperzine-A, Clausenamide et al influence the course of AD obviously. Compound nootropics to improve memory have advantage of low side effect and multiple targets are worth studying. In the study of last ten years,Alzheimer can be prevent and treat by compound nootropics in many pathways,either its deterioration of the memory and histopathology changes in the brain of experimental animal model or hypomnesia of human being can be improved by compound nootropics.Objective To study the screening methods of core herbal combinations of anti-Alzheimer prescriptions by Data Mining Methods and To prove the feasibility of the reverse method studying on Alzheimer in the field of TCM theoretical research.Methods The search keywords of Alzheimer's symptoms were work out, The traditional medical prescriptions for anti-Alzheimer in Dictionary of Chinese Medicine Prescriptions( DCMP) were selected and be normatively approached.TCMIS V2. 5 was used to input the prescription information and construct the database. The commonly used herbs,combination principles,Dominance & Recessive herb pairs and the correlations between prescriptions and symptoms were analyzed and similar &new prescriptions were developed with data mining methods such as association rules,improved mutual information method and complex system entropy clustering. Therapeutic theory of traditional Chinese medicine were researched according to the the reverse relationship between principle,method,recipe and medicines.Results 102 typical anti-Alzheimer prescriptions were concluded based on manual screening. 30 commonly used herbs were concluded based on the statistic frequency analysis of 102 anti-Alzheimer prescriptions. Ginseng Radix et Rhizoma, Polygalae Radix, Acori Poria and Tatarinowii Rhizoma ranked top four in frequency. The ingredient of anti-Alzheimer prescriptions were sweet &warm tonic and most of it owe xin & shen channel tropism. 20 commonly used herb pairs were concluded based on the association rules analysis of 102 anti-Alzheimer prescriptions. Ginseng Radix et Rhizoma-Polygalae Radix, Polygalae Radix-"Tatarinowii Rhizoma, and Ginseng Radix et Rhizoma and Tatarinowii Rhizoma ranked top three among the commonly used herb pairs. In accordance with the correlations between prescriptions and symptoms, the core herbal combinations for anti-Alzheimer prescriptions in DCMP were greatly correlated with Ginseng Radix et Rhizoma, Polygalae Radix, Tatarinowii Rhizoma and Acori Poria. 19 similar prescriptions were concluded based on analysis of similar prescriptions. The proportion between similar herbs of Core Herbal Combinations and similar prescriptions were concluded based on manual conversion. 40 new core combinations and 2 pairs of herb combinations for clustering of new prescriptions and 2 newly developed prescriptions were concluded based on improved mutual information method and complex system entropy clustering in 102 prescriptions for anti-Alzheimer. The core pattern of syndrome,core pathogenesis and core therapy of Alzheimer were concluded according to the the reverse relationship between principle,method,recipe and medicines.Conclusion The ingredients in core combinations of anti-Alzheimer prescriptions were highly coincided with those in Kaixin San in DCMP,with clear treatment significance for memory loss and great medical interest. The two newly developed prescriptions for anti- Alzheimer found by TCMIS V2. 5 have potential clinical value. The reverse method studying on Alzheimer in the field of TCM theoretical research is feasible.
