Objective:We extracted, purified and identified the sesquiterpenoid compound of Carotae Fructus. We studied the antioxidant activity and mechanism of the compound preliminarily, which provided a scientific basis for natural antioxidants exploitation.Methods:Adopting reflux extraction, solvent extraction and silica gel column chromatography technique, we extracted,seperated and purified the sesquiterpenoid compound of Carotae Fructus. The chemical reaction and spectral data method were employed to identify the structure of the compound. We studide cell toxicity, antioxidant activity and protein expression of the new compound with the methods of DPPH radical scavenging, FRAP, Western-Blot, MTT experiment and the determination of T-SOD, GSH, MDA.Results:The new compound was elucidated as 4?,9,11,11-tetramethyl-9-hydroxy-7,11-epoxy-5?,10?-naphthalie-1-O-?-D-glucoside. The IC50 of the DPPH radical scavenging was 192.66?mol/L(vitamin C,108.27?mol/L). In order of most antioxidant capacity, it goes: vitamin C, 400?mol/L, 200?mol/L, 100?mol/L, 50?mol/L, and 25?mol/L of the new compound.The cell vitality of L929 cell lines treated with the new compound were close to 100%. The new compound not only strengthened cell vitality,but also increased intracellular GSH and T-SOD content while L929 cell lines experienced oxidative stress injury. The new compound weakened the PARP protein expression of the experienced oxidative stress cells, meantime, it strengthened PI3 K,Akt,m TOR protein expression and Akt,m TOR phosphorylation degree.Conclusion:The new compound has strong DPPH radical scavenging ability and FRAP antioxidant capacity. Meanwhile, the new compound has a protective effect for oxidative stress damage L929 cell lines, which has no significant toxity on the cells. The leakage reduction of T-SOD and GSH from cells reflect the protection on the oxidative stress damage L929 cell lines. The protection also embodys in the reduction of PARP protein expression and the activation PI3K/Akt/m TOR signal path, which has high application value for natural antioxidant.
