Objective: This paper aims to study the chemical constituents from the roots of Lonicera japonica, and the isolated monomer compounds were tested for anti-inflammatory and in vitro anti-viral activities, in order to provide reference for further comprehensive development and utilization of Lonicera japonica resource.Method: The dried-up, powdered roots of Lonicera japonica were extracted with 95 % ethanol under reflux for three times, and then extracted successively with petroleum ether(PE, 60~90 oC), Et OAc, and n-Bu OH. Compounds were isolated and purified by recrystallization and chromatography on silica gel column, Sephadex LH-20 column, semipreparative HPLC etc.. Their structures were elucidated by means of MS, 1H-NMR, 13C-NMR and so on. Part of the isolated compounds were tested for their anti-inflammatory activities against zebrafish model and in vitro antiviral activity against neuraminidase.Result: Thirty-one compounds were isolated and purified from the petroleum ether part and the ethyl acetate part of the roots of Lonicera japonica through chromatographic separation, and identified by a variety of spectrum methods. They were elucidated as chrysophanol(1), palmarumycin CP2(2), ?-sitosterol(3), stigmasterol(4), stigmast-4,6,8(14),22-tetraen-3-one(5), erythrinassinate D(6), lanosterin(7), 3-O-acetyloleanolic acid(8), 3,13-dihydroxystemodan-2-one(9), 4-hydroxycinnamic acid(10), 1-oxo-(1H)-cyclopenta[b]benzofuran-7-carbaldehyde(11), protocatechuic aldehyde(12), chlorogenic acid(13), loganin aglycone(14), daucosterol(15), caffeic acid(16),(E)-3-(3,4-dihydroxybenzylidene)-5-(3,4-dihydroxyphenyl)-2(3H)-furanone(17), secologanin dimethyl acetal(18), korolkoside(19), coniferin(20), sweroside(21), lomacarinoside B(22), secoxyloganin(23),(2E,6S)-8-(a-L-arabinopyranosyl-(1 "?6')-?-D-glucopyranosyloxy)-2,6-dimethyloct-2-eno-1,2"-lactone(24), 5-O- caffeoylquinic acid(25), chlorogenic acid methyl ester(26), chlorogenic acid ethyl ester(27), 3,5-O-dicaffeoylquinic acid(28),4,5-O-dicaffeoylquinic acid(29), grandifloroside(30), and 3,4-O-dicaffeoylquinic acid(31), respectively, including two new compounds and eight firstly separated ones from this plant. Anti-inflammatory activities of compounds from the ethyl acetate extract, including 1, 2, 5, 7, 6, 8, 9, 10, 11, 12, 13, 14, 17, 18, 21, 22, 23, 24, 26, 28, 29, 30, and 31, were tested against Zebra fish. It was found that compounds 28 and 29 showed anti-inflammatory activities with the concentration at 1 ?g/m L, 10 ?g/m L, 100 ?g/m L; compound 13 showed anti-inflammatory activity with the concentration at 10 ?g/ml, 100 ?g/m L; compounds 8, 11, 12, 30, and 31 showed anti-inflammatory activities with the concentration at 100 ?g/m L; compound 31 showed promote inflammation with the concentration at 1 ?g/m L; compound 22 showed promote inflammation with the concentration at 100 ?g/m L; antiviral activities of compounds 1, 2, 5, 6, 7, 8, 9, 10, 11, 12, 13, 14, 16, 18, 19, 21, 22, 23, 24, 25, 26, 27, 28, 29, 30, 31 were tested against neuraminidase. It was found that compounds 2, 5, 8, 11, 12, 13, 24, 25, 27, 28, 29, 30, and 31 showed antiviral activities with the concentration at 1 mg/m L.Conclusion: The reserach of the petroleum ether and the ethyl acetate parts of the roots of Lonicera japonica got 31 compounds. Through the study of the anti-inflammatory activities and the antiviral activities of these compounds, it will provide reference for further comprehensive development and utilization of this title plant.
