Objective: Renal cell carcinoma is the most lethal urological malignancy. In our previous study, using microarray analysis to study expression profiling of micro RNAs, we demonstrated that miR-144-3p was remarkably upregulated in clear cell renal cell carcinoma(cc RCC) tissues compared with matched normal renal tissues. This study aimed to identify the clinical significance of miR-144-3p and its influence on tumor growth of clear cell renal cell carcinoma.Methods: Relative miR-144-3p expression levels in cc RCC tissues were analyzed by q RT-PCR. We injected 1×106 SN12-PM6 cells infected with lentiviral particles carrying miR-144-3p and miR-NC into the flanks of 5-week-old nude mice and then tumor growth was evaluated. Statistical comparisons between groups were analyzed and a P-value <0.05 was considered to indicate a statistically significant difference.Results: A total of 110 cases of clear cell renal cell carcinoma tissues were analyzed by q RT-PCR. The miR-144-3p expression level was significantly increased in T3+T4 group compare with T1+T2 group, but no statistically significant difference was found in age, gender and tumor Fuhrman grade. Tumor formation rate, tumor volume and tumor weight of miR-144-3p group was significantly higher than of the control group.Conclusions: MiR-144-3p expression level is linked to tumor TNM stage, but not associated with age, gender or tumor Fuhrman grade. Mi R-144-3p promotes kidney tumor formation and growth and may play an important role in the pathogenesis of renal cell carcinoma.
