Objectives To observe the effects of Shizidaiping Formula on blood glucose and lipid metabolism and adipose tissue IR-related proteins in spontaneously diabetic GK rats, and preliminary dicussion its action mechanism of IR improvement in T2 DM.Methods In this study of spontaneous type 2 diabetic GK rat as model, Model rats were randomly divided into 5 groups: the model group, Shizidaiping Formula groups of low, middle and high, the Metformin group, Each group 8(n=8). Another 8 Wistar rats were randomly selected as normal control of(Control). The rats were fed the appropriate feed for 8 weeks. During the test, Rats were observed daily water intake, food intake and general state; body weight was weighed weekly; take the tail vein of a very small amount of blood, random blood glucose test strip to detect fasting blood glucose, postprandial blood glucose two hours and the oral glucose tolerance tests were performed and analyzed before and after the drug administration. At the end of the 8 week, aortic blood was collected for the measurement of serum total cholesterol(TC), triglycerides(TG), high density lipoprotein(HDL), low density lipoprotein(LDL). ELISA kit tset fasting insulin, the C-peptide levels by radioimmunoassay. Clipped adipose tissue cryopreserved for inspection; Immunoblotting to detect adipose tissue glucose transporter 4(GLUT-4), insulin receptor substrate 1(IRS-1), peroxisome proliferator-activated receptor(PPAR?) expression of protein.Results 1 Normal Wistar rats in good condition, shiny coat, active.Compared with the normal group, model group GK rat grows weeks of age with varying degrees of polydipsia, polyuria, rough coat, spiritual malaise, slow response and other symptoms; With the advance of treatment time, compared with the model group, Shizidaiping Formula group rats of the above were significantly improved. 2 Compared with the normal group, model group increased fasting,postprandial 2h blood glucose levels and insulin resistance index(P<0.05); compared with the model group, Shizidaiping Formula of rats reduced fasting and postprandial 2h blood glucose levels, and insulin resistance index decreased(P<0.05). 3 Compared with normal group, model group rats total cholesterol, triglycerides, low-density lipoprotein levels were significantly increased; Compared with the model group, Shizidaiping Formula reduces total cholesterol, triglycerides, low-density lipoprotein levels, thereby improving the degree of lipid disorders. 4 Fasting insulin levels were no significant differences in Shizidaiping Formula of fasting insulin had no significant effect; compared with the normal group, all C-peptide levels were significantly lower, SZDPF-H group which have no significant differences in C-peptide levels with the normal group. 5 Compared with the normal group, the adipose tissue of rat model group PPAR?, Glut-4, IRS-1 protein was significantly decreased(P<0.05); Compared with the model group, Shizidaiping Formula group's Glut-4, PPAR?, IRS-1 protein content were increased(P<0.05).Conclusions 1 Shizidaiping Formula can reduce spontaneously diabetic GK rats fasting plasma glucose and postprandial blood glucose, improve glucose tolerance in GK rats. 2 Shizidaiping Formula can regulate blood lipids, improve blood lipid level disorders. 3 Shizidaiping Formula can improve IR T2 DM rats, which may be related to the increasing of PPAR?, Glut-4, IRS-1 protein level in adipose tissue.
