ObjectiveBy detecting serum 8-hydroxy-2-deoxyguanosine(8-OHd G) levels and its relevant gene- human 8-oxoguanine glycosylase 1( h OGG1) gene polymorphisms in patients with coronary artery ectasia(CAE), to explore the potential association of 8-OHd G level and human 8-hydroxyguanine glycosylase( h OGG1) gene polymorphism with CAE.MethodsThis study included 168 patients who underwent coronary angiography in Tianjin First Central Hospital during Nov 2014 to Oct 2015, including 59 patients with CAE,56 patients with CAD and 53 patients with normal coronary arteries. All the patients' following clinical data was recorded, such as gender, age, hypertension history,diabetes history, smoking and drinking history, total cholesterol, etc. Serum 8-OHd G levels were measured by enzyme linked immunsorbent assay(ELISA) method, then comparing the differences between three groups and investigating the relation between 8-OHd G level and CAE by using logistic regression. The h OGG1 gene polymorphism was detected by polymerase chain reaction- restriction fragment length polymorphism(PCR-RFLP) methods, then comparing the differences between three groups. The relation between the h OGG1 gene polymorphism and CAE was investigated by using logistic regression.Results:1. Serum 8-OHd G levels of three groups didn't have statistical differences. But after comparing serum 8-OHd G levels of each two groups, we found that serum8-OHd G levels had statistical differences between CAE group and NCA group(CAE group:98.33±30.98ng/m L, NCA group:86.36±25.95ng/m L, p=0.029<0.05). And there weren't statistical differences between CAD group and NCA group( p>0.05).2. Univariate logistic regression analysis to serum 8-OHd G level revealed serum8-OHd G level maybe a risk factor for the development of CAE(OR=1.019,95%CI=1.001-1.038, p<0.05). In order to get rid of the influences of gender, age,hypertention and diabetes, multiple logistic regression analysis to serum 8-OHd Glevel was proceed. The result showed that the relationship between serum 8-OHd G level and the development of CAE had statistically significance(OR=1.023,95%CI=1.000-1.047, p=0.049<0.05).3. The analysis of genotypes and alleles showed that: the most common type of genotypes was Ser/Ser, and Cys/Cys genotype was the fewest. But there weren't statistical differences of genotypes and alleles in patients between three groups(p>0.05).4. Univariate logistic regression analysis to h OGG1 Ser326 Cys gene polymorphism revealed that the genotypes(Ser/Ser genotype, Ser/Cys genotype and Cys/Cys genotype) didn't have statistically significant correlation with the development of CAE(p>0.05). After adjustment of gender, age, hypertention and diabetes, etc, multiple logistic regression analysis to h OGG1 Ser326 Cys gene polymorphism was proceed. The result showed that it still didn't have statistically significant correlation between the genotypes and CAE.5. The comparison of serum 8-OHd G levels of the genotypes in three groups showed that serum 8-OHd G levels of(Ser/Cys genotype+ Cys/Cys genotype) were higher than that of Ser/Ser genotype, and the difference was statistical significant(p<0.05).Conclusions:1. The elevation of serum 8-OHd G level may be a risk factor for the development of CAE.2. The h OGG1 Ser326 Cys gene polymorphism didn't have statistically significant correlation with the development of CAE.
