Diabetes is a metabolic disorder syndrome the cause of which is complicated and the complications from head to toe. The characteristics of the high incidence and high morbidity makes it become a research hotspot. Onder the high glucose condition, blood viscosity increased effective and result in blood vessel function disorder, cause a variety of chronic complications and vascular endothelial damage is known as the initiating factor of cardial-cerebrovascular disease. Hawthorn can be called Chinese endemic species, leaves or fuits as the medicine, hawthorn leaf extract used in treatment of the prevent cardialcerebrovas-cular disease have a Significant effect such as coronary heart disease,hyperlipidemia and blood-vessel. Animal experiments show that hawthorn leaf flavonoids have an intervention effect on plasma glucose and complications of diabetes on animal model, but the mechanism remains to be further research.The raw material quality identification and separation were the main monomer in this paper. means of the cell biology were used in research of in vitro hypoglycemic activity of rhamnosylvitexin and glycoside in order to explore the hypoglycemic mechanism of the hawthorn leaves flavonoids. The main contents as follows:1 The raw materials quality of four main producing areas in China were identified,they were Shandong,Hebei,Shanxi and Liaoning. Hebei Yan red petals and Shandong Venus were used as the preferred base. Total flavone glycosides were measured by using acid hydrolysis-HPLC, in order to characterization of total flavonoids.2 Through the test in the laboratory to industrial production, extraction method and purificatio-n method of industrial production of total flavonoids was optimized,the most optimal solutio-n is:70% methanol extract four times, each time 1.5h and the tempreture must be 70?,eight-fold and three times of six-fold solvent were used respectively compared with the inventory.One ponint of five-fold of water was entered after The solvent were recycled until the extract became Viscously.D101 macroporous resin purification were used to purification,3 times column volume 10%methanol were used to removal onder the flowrate of 1BV/h and 90% methanol used to enrich slower the flowrate of 2BV/h;The target compounds Rhamnosylvitex-in were isolated from hawthorn leaves flavonoids.3 Means of of cell biology were used to research for the effect of Rhamnosylvitexin and glycoside on oxidatxive stress in human umbilical vascular endothelial cell induced by highgl-ucose,and research the pathogenesis of that.results show that:Rhamnosylvitexin which add in onder the 10'5 M and hatch less than 72 h have no effect on normol human umbilical vascular endothelial cell.10-9M,10-8M,10-7 M Rhamnosylvitexin and 10-8M glycoside can resdused the content of MDA(P<0.01) and increased viability of SOD (P<0.05) significantly,glycoside was beter than others. Rhamnosylvitexin have an Antagonism cellapoptosis activity through promote the gene and protein expression of Bcl-2 and reduced the gene and proteinexpression of Caspase-3..Through the previous Rhamnosylvitexin and glycoside can anti the oxidized stress damage induced by high glucose. glycoside was more excellent in those filed.
