Study On The Chemical Constituents In Four Zhong-jing-fang Formulae

Dang-Gui-Shao-Yao-San(DSS),Zhi-Zi-Da-Huang-Tang(ZZDHT),Yin-Chen-Hao-Tang(YCHT),and Da-Huang-Xiao-Shi-Tang(DHXST)were four commonly used formulae from Zhong-Jing-Fang.Until now,the literature about the study of chemical constituents in the four formulae is rare.Angelicae sinensis radix,monarch drug in DSS,is a widely used crude drug in Zhong-Jing-Fang.The separation and study of chemical constituents in Angelicae sinensis radix is of great importance in the further study of formulae that contains Angelicae sinensis radix.In this paper,six compounds were isolated from Angelicae sinensis radix,including a novel one.To study the chemical consituents in DSS,LC-Q-TOF-MS and LC-IT-MS was used to analyze 70%ethanol extract from DDS.30 compounds in 70%ethanol extract from DDS could be identified.And,fourteen compounds were identified for the first time in DSS,including organic acids,protostane trierpenoids,cyclopeptides,and other types of compounds.This work dissected the chemical constituents of DSS,and the results could provide evidence for the research on effective constituents and quality control of DSS.LC-Q-TOF-MS and LC-IT-MS in positive and negative mode were applied to identify and compare the constituents in ZZDHT,YCHT,and DHXST.A total number of 112 components were detected in these three formulae.And,based on the elemental composition of constituents given by LC-Q-TOF-MS and their multi-stage MS fragmentation information provided by LC-IT-MS,89 constituents were identified,including organic acids,terpenoids,alkaloids,flavonoids,coumarins,cyclopeptides,stilbene glucosides,anthraquinone,and other types of compounds.
