Background: Alcoholic liver injury is a common cause of liver cirrhosis,which is caused by abundant alcohol for long term.Furthermore,alcoholic liver disease(ALD)has been noticed gradually.The incidence of alcoholic liver injury is increased gradually,only following the Viral hepatitis in China.ALD is very harmful to the health of human,with high incidence and severe effects on nervous system and reproductive system[1].Liver cirrhosis and liver cancer occurs easily when ALD co-exists with hepatitis B,hepatitis C.Therefore,people should pay more attention on the prevention and cure of ALD.The prevention and treatment of ALD has become the important task at present.There are many studies about the effects of radix linderae on the treatment of digestive diseases,testicular diseases,h Cardiovascular diseases and rheumatic disease while few studies are found about effects of radix linderae on the treatment of liver diseases.A few studies have shown that radix linderae could inhibit the elevated levels of serum transaminases induced by CCl4 or ethionine and could prevent liver from fatty infiltration.Furthermore,furan sesquiterpene was considered to be the effective ingredient of radix linderae to protect liver injuries.However,only the early extracts of radix linderae are studied,the further effective extracts of radix linderae are not studied until now.Aims: To investigate the effects of different extracts of radix linderae of tian tai on acute alcoholic liver injury in rats.Methods: The model of acute alcoholic liver injury was induced by different concentrations of liquor and different extracts of radix linder orally for 10 days in SD rats.Serum levels of total cholesterol(TC),triglyceride(TG),alanine aminotransferase(ALT)and aspartate aminotransferase(AST)were determined by chemical methods.The activities of superoxide dismutase(SOD)andmalondialdehyde(MDA)levels in serum and liver homogenate were determined by an spectrophotometer.The pathological change of liver was determined by HE staining.The expressions of NF-?B,TNF-? and IL-1? in liver were determined by immunohistochemistry.The expressions of CYP2E1 m RNA in liver were determined by real time polymerase chain reaction(RT-PCR).Results: The ethanolic extracts of radix linderae reduced serum ALT levels significantly compared to the model group(P<0.05).The ethanolic extracts and the extracts of petroleum ether of radix linderae reduced serum AST levels compared to the model group(P<0.05).serum TC levels were decreased by the water extracts,ethanolic extracts and the extracts of petroleum ether of radix linderae and water layer of radix linderae after extracting compared to the model group(P<0.05).The ethanolic extracts and the N-butyl alcohol extracts of radix linderae decreased serum TG levels significantly compared to the model group(P<0.05).The ethanolic extracts of radix linderae and water layer of radix linderae after extracting reduced serum MDA levels compared to the model group(P<0.05).The SOD activities in serum and liver were increased by the water extracts,the ethanolic extracts,the N-butyl alcohol extracts of radix linderae and the water layer of radix linderae after extracting compared to the model group(P<0.05).Different extracts of radix linderae attenuated the pathology of liver compared to the model group.Different extracts of radix linderae reduced the expressions of IL-1?,NF-?B,TNF-? and CYP2E1 m RNA in liver compared to the model group(P<0.01).Conclusion: Different extracts of radix linderae protected acute alcoholic liver injury.Compared to other extracts of radix linderae,the protective effects were better of alcohol extracts,the extracts of petroleum ether and the water layer of radix linderae after extracting.The mechanisms underlying the protective effects of different extracts of radix linderae were related to its antioxidant effects,its inhibition of the expressions of IL-1?,NF-?B,TNF-? and CYP2E1 m RNA in liver.
