Objectives: 1.By the method of dietltylmitrosainine-induced hepatocellular precancerous lesion model,investigate its feasibility and reliability of the precancerous lesions;2.Through the study of DENA-induced hepatocellular precancerous lesion model,it is helpful to study its effect about liver function and the mechanism of compound Yipi Yanggan decoction on the formation of inflammatory micro-environment during the process of liver precancerous lesion in rats.Methods: Adaptive feeding 1 week before the experiment,SPF grade wistar rats who were 80 males were randomly divided into blank group,model group,low-dose and high-dose group of compound Yipi Yanggan decoction,whose quantities were 20 in every group.At the beginning of the experiment,DENA(0.5ml/100 g,2times/ week(once/84h),50mg/kg/week),was continuously injected into enterocoelia to 16 weeks for inducing liver precancerous lesions model.At the same time,the rats in the low-dose group were treated with compound Yipi Yanggan decoction(0.5g/ml,10ml/kg,once/d)and the high-dose group treating with the compound(1.0g/ml,10ml/kg,once/d),they were continuously intragastric administration to 16 weeks during the process of liver precancerous lesion in rats.The rats were sacrificed at the 16 th week after 24 hours of intragastric administration.The rats were weighed before they were sacrificed.After they died,the blood was collected,and the liver and spleen were removed and weighed,and the survival rate,the incidence of liver cancer,the liver and spleen index were calculated.Detect the changes of liver function index about AST,ALT,GGT,ALP and TBIL.Hematoxylin-eosin staining(HE staining)was used to observe the pathological changes of the liver.Measure the levels of Toll-like receptor 4(TLR4)?and nuclear factor ?B(NF-?B)in liver tissue and interleukin-6(IL-6),tumor necrosis factor-?(TNF-?)in serum.Results:1.Compound Yipi Yanggan decoction on the survival rate of DENA-induced hepatocellular precancerous lesion model was significantly improved.The model group,low-dose and high-dose group survival rates of compound Yipi Yanggan decoction were 85%,90%,95%(P> 0.05).Meanwhile,the incidence of liver cancer was decreased,the incidence rate was 10%,5% and 0%,respectively.There was no significant difference between the two groups(P> 0.05),and the liver and spleen index were decresed(P <0.05).The body weight of the rats in each group increased from the model group,but the increase rate was the highest in the blank group,followed by compound Yipi Yanggan high-dose group and low-dose group,the model group(P> 0.05).Until the death of rats,the comparison of body weight changes found: the body weight of the rats in the high-dose group which compare with the model group was increased insignificantly(P> 0.05).2.Compound Yipi Yanggan decoction had a significant effect on DENA-induced liver function in rats with hepatocellular precancerous lesions: Compared with the blank group,the liver function of the model group decreased significantly,but the changes of liver function index about AST,ALT,GGT,ALP,TBIL were significantly increased(P <0.01).Compared with the model group,the low-dose group about ALT?AST?GGT?TBIL decreased obviously(P <0.05),but the improvement of ALP was not(P> 0.05),but the high-dose group about AST,ALT,GGT,ALP,TBIL were significantly decreased(P <0.01).the comparison of each group of compound Yipi Yanggan decoction about liver function index about AST,ALT,GGT,ALP,TBIL were significant(P <0.01).It is suggested that the high-dose group is more significant benefits,which is compared with low-dose group.3.Compound Yipi Yanggan decoction could decrease the expression of IL-6 and TNF-?in serum,TLR4 and NF-?B in liver tissue in the process of DENA-induced hepatocellular precancerous lesions: compared with the blank group,the expression of inflammatory factors IL-6,TNF-?,TLR4 and NF-?B in the serum and liver tissues were significantly higher(P <0.01).Compared with the model group,the small-dose group about TNF-?,TLR4 and NF-?B decreased obviously(P <0.05,P <0.05,P <0.01),but the expression of IL-6 did not show a significant regulatory role(P> 0.05);and the high-dose group about IL-6,TNF-?,TLR4 and NF-?B were significantly decreased(P <0.01).the comparison of each group of compound Yipi Yanggan decoction found:the changes of TNF-?,IL-6,TLR4 and NF-?B were significantly higher in high-dose group(P <0.01).Conclusion: 1.It is simple,easy to operate,low mortality rate and low canceration rate,which can be used as a good method by the DENA-induced rat liver precancerous lesion model.2.Compound Yipi Yanggan decoction can significantly improve the liver function of DENA-induced hepatocellular carcinoma model rats.3.Compound Yipi Yanggan decoction can inhibit the formation of inflammatory micro-environment by reducing the level of inflamematory factors in the liver of rats with precancerous lesions.4.Compound Yipi Yanggan decoction about the formation of precancerous lesions and the progress of liver cancer having an important inhibitory effect is expected to become an important means of Chinese medicine to prevent liver cancer.
