Objective:To study the effect of Yipi Yanggan decoction(YPYGF)on the amel ioration of pathological morphology and expression of NF-?B?STAT3 in dieth ylnitrosamine(DEN)induced rat liver precancerous lesion and to explore the pr evention and treatment methods of traditional Chinese medicine in hepatocellular carcinoma.Methods:Adaptive feeding 1 week before the experiment,SPF grade wistar rats who were 70 males were randomly divided into blank group,model group,lo w-dose and high-dose group of compound Yipi Yanggan decoction.At the beginning of the experiment,DEN was continuously injected into enterocoelia to 16 weeks for inducing liver precancerous lesions model.At the same time,the rats in the low-dose group were treated with compound Yipi Yanggan decoction(0.5g/ml,10ml/kg,once/d)and the high-dose group treatingwith the compound(1.0g/ml,10ml/kg,once/d),they were continuouslyintragastric administration to16 weeks during the process of liverprecancerous lesion in rats.The rats were sacrificed at the 16 th week after24 hours of intragastric administration.The rats were weighed before they were sacrificed.After they died,the blood was collec ted,and the liver was removed.Observe rat general situation and the weight.Detect the changes of liver function index about AST,ALT,GGT,ALP and TBIL.Hematoxylin-eosin staining(HE staining)was used to observe the pathological changes of the liver.the expressions of NF-?B and STAT3 of liver tissues were measured by immunohistochemical detection.Results:1.Compound Yipi Yanggan decoction on the survival rate of DEN-induc ed hepatocellular precancerous lesion model was significantly improved.The model group,low-dose and high-dose group survival rates of compound Yipi Ya nggan decoction were 80%,90%,95%(P>0.05).The body weight of the rats in each group increased from the model group,but the increase rate was the highest in the blank group,followed by compound Yipi Yanggan high-dose group and low-dose group,the model group(P>0.05).Until the death of rats,the comparison of body weight changes found: the body weight of the rats in the high-dose group which compare with the model group was increased significant ly(P<0.05).2.Compound Yipi Yanggan decoction had a significant effect on DEN-induced liver function in rats with hepatocellular precancerous lesions: Comp ared with the blank group,the liver function of the model group decreased sig nificantly,but the changes of liver function index about AST?ALT?GGT?ALP?TBIL were significantly increased.Compared with the model group,the lowdose group and the high-group about ALT?AST?ALP?GGT?TBIL decrease d obviously,the liver function decreased significantly(P<0.05).3.In the blank group rat liver tissue,a small number of liver cells showed positive expression.The model group NF-?B and STAT3 were both highly expressed,mainly in t he precancerous lesion and surrounding tissues,showing the nucleus dark brown staining as positive cells.However,both Yipi Yanggan decoction high-dose group and low-dose group can significantly inhibit the expression of NF-?B and STAT3,and the high-dose group has the best effect(P<0.01).Conclusion:1.Yipi Yanggan decoction can significantly improve the liver function and liver function of DEN induced liver cancer precancerous lesion model.2.Yipi Yanggan decoction of pathological tissue in rat liver precancerous lesions form has certain influence,hyperplasia of cell atypia significantly smaller,dualcore cells decreased,and inflammation is lighter,and the effect of high-doseg roup is better.3.The results suggest that Yipi Yanggan decoction significantly suppressprecancerous lesion of rats' liver by mediating expressions of NF-?B and STAT3.
