Objective This study mainly discuss the probablely mechanism of the selenium-rich earthworm to intervene the hepatic fibrosis of mice,this can provide theoretica basis for the selenium riched earthworm to cure hepatic fibrosis.Method Making the Male Kunming mice 60 randomized divided into four groups: the blank group,the model group,the experimental group,The control group,each group of 15.expect the blank group,the other three groups mice were given subcutaneous injection with CCl4?carbon tetrachloride olive oil solution?of 30%,1 m L/kg every two days building the model.At the same time the mice in the model group were gavage intervention with giving injection with physiological saline of 0.9% every day one time.The mice in the blank group were gavage intervention with giving injection with physiological saline of 0.9% every day one time.The mice in the the experimental group were gavage intervention with giving injection with the suspension of selenium-rich earthworm every day one time.The mice in the control group were gavage intervention with giving injection with the suspension of ordinary earthworm every day one time.stop experiment building After 12 weeks,and continue gavageing intervention about 2 weeks.After the last gavage,fasting for 12 hours and kill the mice.This study use the HE staining and Sirius Red staining learn about the liver pathologic changes.Microplate method to detect the ALT,AST,Urea enzyme test mice serum BUN content,amino acid oxidase method to detect serum Scr.The Hyaluronic acid and Laminin were measured by ELISA.The PCIII,COV-IV and TGF-?1 were measured by immunohistochemical method.Recording the mice tail bleeding time and clotting time,Fully automatic blood rheometer detection the whole blood viscosity and the plasma viscosity.Compare each group mice the weight change,the coefficient of liver,the coefficient of spleen and the coefficient of kidney.Result The levels of ALT,AST,HA and LN in the serum of the model group were higher than the other three group.The blank group and the model group compared have statistical significance?p<0.01?.The coefficient of liver and the coefficient of spleen results show that the model group was higher than the other three group,the numerical level of selenium-rich earthworm group close to the blank group.the BUN,Scr of serum and the coefficient of the kidney in the model group are higher than the other three group,have statistical significance?p<0.05?,the express of PCIII,COL-IV and TGF-?1 in the model group are more obvious than the other three group.the blank group and the model group have statistical significance?p<0.01?.hemorrheology results shows: Model group bleeding time,clotting time were lower than the other three group,the whole blood viscosity and the plasma viscosity were higher than the other three group,the blank group and the model group compared with very statistical significance.The weight of each group in the different stage show that there was no big difference between the blank group and the model group when the fourth week,there is big difference between the selenium-rich earthworm group and the blank contrast group,so is the ordinary earthworm group,and have statistical significance?p<0.05?.from the fourth week,the model group mice weight lower than other groups,the model group compared the blank contrast group has statistical significance?p<0.05?.Conclusion The selenium-rich earthworm can reduce the TGF-?1,HA,LN,PCIII,COL-IV of hepatic tissue,improve the liver function,reduce ALT and AST vitality.Changing the parameters of hemorrheology,reduce the coefficient of liver and the coefficient of spleen.relieve the blood stasis of the liver and spleen.Increase the weight and improve the nutritional status.Through the kidney security evaluation,found that selenium-rich earthworm has protective effect to the kidney.To sum up the selenium-rich earthworm has effective intervention to the hepatic fibrosis of the mouse induced by CCl4,and probablely through TGF-? signal channel.at the same time have no-toxic and side effects on kidney to the hepatic fibrosis of the mouse.
