Objective:To investigate the effects of selenium-rich earthworm's solution on the expression of BMP-2 and VEGF in the left femoral fracture healing of SD rats,explore the effect of selenium-rich earthworm's solution on fracture healing and its possible mechanism.Methods :72 male SD rats were selected to make the left femoral shaft fracture model.Then they were divided into three groups: the Shangke Jiegu group(A),the selenium-rich group(B),the normal saline group(C),each group included 24 SD rats.Later,gave respectively Shangke Jiegu Tablet 300mg/(kg·d),selenium-rich earthworm's solution 300mg/(kg·d),equal volume of saline.At 2nd,4th and 6th week after fracture operation,8 rats in each group were tested by observation of the gross specimen,callus volume measurement,DR radiography and HE staining,and detected the expression of BMP-2 and VEGF.Finally,the results of each group were analyzed by SPSS 20.0.Results:1.Fracture imaging: At 2nd week after fracture,a small amount of callus was found in each group,and the fracture line is clear.At 4th week,most of the specimens of each group had formed bone callus,fracture line became blurred.At 6th week,each group reached healing criteria.There was no significant difference between the two groups at the same period(P> 0.05).2.Specimen observation and callus measurement: At 2nd week after fracture,evident callus formation was observed in each group,its volume is big,and texture slightly soft.At 4th week,the volume was smaller and the texture became harder.At 6th week,the volume was further reduced and changed,and there was no difference by eyes.In addition,to measure callus' volume.By statistical treatment,there was significant difference between A and B group,A and C group(P<0.05),and others' no significant difference between the two groups at the same period(P> 0.05).3.Callus histological observation: At 2nd week after fracture,bone trabeculae was found in endosteum and periosteum of each group both ends of the fracture line,and fracture ends were jointed by fibrous tissue,the amount of bone in the membrane and cartilage island area of A and B groups were significantly larger than C group.At 4th week,trabecular bone of each group further increased,and connected the two fracture ends.At 6th week,two bone fracture ends of the trabecular bone completely transformed into lamellar bone,medullary cavity recanalization in A and B groups.Trabecular bone of C group was gradually transited to the lamellar bone,part of the specimen cavity recanalization.Statistical analysis was statistically significant.4.The expression of BMP-2: The positive expression of BMP-2 was found in fibroblasts,chondrocytes and osteoblasts of callus tissues.At first,it mainly expressed in osteoblasts,bone cells in trabecular bone and near the callus outer part of the naive chondrocytes.And in late stage,mainly expressed in osteoblasts.At 2nd,4th,6th weeks,A and B groups' Cytoplasmic staining depth was significantly greater than C group.The MOD value of BMP-2 positive expression in each group was the highest at 2nd week after fracture,and decreased gradually at 4th week,the most significant decline in 6 weeks and in the lowest expression.By statistical treatment,at 2nd week,there was significant difference between A and B group,A and C groups(P <0.05);at 4th,6th weeks,significant difference between A and C groups,B and C groups(P <0.05).5.The expression of VEGF: A and B groups were respectively significantly higher than C group in peripheral serum(P <0.05).Conclusion:The selenium-rich earthworm's solution can promote the expression of VEGF in callus tissue and BMP-2 in serum.Thereby to promote fracture healing.
