Objective: Osteoporosis is a disease characterized by the reduced of bone mass and deteriorate bone microstructure,which decreaseds bone strength,increased skeleton fragility and the risk of fracture.The employ of exercise in the treatment and prevention of osteoporosis can effectively reduce the family economic burden and the side effort caused by drug treatment.Exercise play a key role in the prevention and treatment of osteoporosis by mechanical stress stimulation,cytokines and bone metabolism signal pathway.Recent studies have shown that non coding RNA plays an important role in bone metabolism.The purpose of this study was to establish a osteoporosis model of mouse and trained with moderate intensity tredmill exercise to explore the effect of exercise on bone long non encoding RNA(long noncoding RNA,lncRNA)of the osteoporosis mouse whic would provide a basis theory to define the action mechanism.Methods: Forty-four 8 weeks old female C57BL/6 mices were randomly divided into to sham operation group(SHAM),sham operation + exercise group(SHAM+EX),ovarian group(OVX),OVX + exercise group(OVX+EX);a 1cm incision on the back of paravertebral,length 1/3 to ovary,sham operated mice incision at the same part of the removal of the same size of fat.4 weeks after surgery,SHAM+EX group and OVX+EX group conduct treadmill training of 25 degrees for 9 weeks,5 days a week and rest at Saturday and Sunday.The first week of exercise preconditioning,speed 6m/min for 30min;and then started speed of 8m/min,an increase of 1m/min per week;the training conclude 5 min warm-up exercise(speed: 6m/min)and 55 min formal training.Group SHAM and group OVX were not subjected to any intervention.After the end of intervention from hematuria and left and right femur tibia.Measure bone density,bone biomechanics and mcro CT index of the right femur and the bone histomorphometry of the left femur.selected 3 mice in each group to test the expression of the lncRNA.The data were analyzed by software SPSS 20.Results:(1)Ovariectomy can significantly increase the body weight of mice,and exercise can reduce the weight gain of mice due to ovarian removal.(2)Effect of exercise on bone mineral density(BMD): Exercise can significantly improve the BMD of sham operated group and osteoporosis group,compare to the SHAM group: the BMD of the SHAM+EX group was significant increased(P < 0.05)and the OVX group was significanted decline(P < < 0.01).The BMD of the OVX+EX group was significant increased campred to the OVX group(P < 0.01).(3)Effects of exercise on bone histomorphometry in osteoporosis mice: The MAR of SHAM +EX group is higher than the SHAM group,but there is no significant differences.The MAR of SHAM group is significant higher than the OVX group(P < 0.05).The MAR of OVX+EX group is significant higher than the OVX group(P < 0.05).Exercise can improve the sham and osteoporosis in mice BFR/BV but there is no significant difference.Compared with the SHAM group,the OB.S/BS of the SHAM+EX group was significantly increased(P < 0.05),the OVX group was significantly lower(P < 0.01).Compared with SHAM group,the OC.S/BS of OVX group increased significantly(P < 0.01).Compared with OVX group,OC.S/BS in OVX+EX group was significantly lower(P < 0.01).(4)microCT detection: Exercise can significantly improve the BV/TV,Tb.Th and Tb.N in sham operated mice and osteoporosis mice,but there were no significantly difference in the results.(5)The effects of exercise on bone biomechanics in osteoporosis mice: Compared with SHAM group,SHAM+EX group,the maximum load increased significantly(P < 0.05),OVX group decreased significantly(P < 0.05).Compared with OVX group,OVX+EX group,the maximum load increased significantly(P < 0.05).Compared with the SHAM group,the SHAM+EX group had no significant difference in the flexural strength,but significantly decreased in the OVX group(P < 0.05),which was significantly lower than that in the OVX+EX group(P < 0.05)compared with the OVX group.Compared with SHAM,the elastic modulus of SHAM+EX group increased,but there was no significant difference,the elastic modulus of OVX+EX group was higher than that of OVX group,but there was no significant difference.(6)The effects of exercise on bone lncRNAs osteoporosis mice: Screening thedifferent expression in 2 fold and 4 fold change,and P < 0.05 lncRNAs.Compared with SHAM group,SHAM+EX group there are 64 up-regulated genes and 161 downregulated in 2 fold change and 3 up-regulated,3 down regulated in 4 fold change.Compared with SHAM group,in the OVX group there were 37 up-regulated and 231 downregulated in 2 fold change and 25(Gm30783?LOC102639568 ect.)upregulated,17(Gm33272?Gm12678?Gm10248?LOC105243625 ect.)downregulated in 4 fold change.Compared with OVX group,in the OVX+EX group there are 181 downregulated,56 up-regulated in 2 fold change and 3(lnc 105247265 ect.)up-regulated genes and 8(Gm31126?LOC105243692?Gm11651 ect.)downregulated genes are in 4 fold change.(7)The signal pathway of Melanoma signaling pathway,mTOR signal pathway,Toxoplasmosis signal pathway,NF-?B signaling pathway and PI3K-Akt signaling pathway may be regulated by lncRNA by KEGG pathway analysis.Regulation of cell cycle phase transition,vocalization behavior,detection of biotic stimulus and other biological processes,TOR complex,synapse part and other cell components to VEGF-A-activated receptor activity,VEGF-B-activated receptor activity and other molecules may be regulated by lncRNAConclusion: Moderate intensity treadmill exercise can significantly improve the bone density of osteoporosis mice,increse the bone formation rate and bone strength,it can also upgulate or downgulate the expression of bone lncRNAs.
