Inonotus obliquus also called chaga,chopped birch and Siberia,ganoderma lucidum,etc.,is a parasite on birch,elm,birch and so on under the bark of the stocking or cut down on the withered trees after fungus fruiting body,is a kind of medicine and food fungi,anti-inflammatory,antioxidant,anticancer,fall blood sugar,reducing blood fat,prevent tumor immunity effect and so on,has high nutritional value,contain fungal peptide,the element of Fuscoporine,alcohol,polysaccharide of Fuscoporine,various antioxidant triterpene compounds and lignin compounds such as a variety of active ingredients.In recent years,as a popular health care product,it has attracted wide attention from scholars.In order to further explore the antioxidant properties and hypoglycemic and lipid-lowering abilities of water extracts from Inonotus obliquus,the following studies were conducted on Inonotus obliquus:In order to explore the best method for extraction of Inonotus obliquus by hot water ultrasonic extraction,the effects of extraction temperature,extraction time and the ratio of material to liquid,i.e.,polysaccharide yield,were investigated respectively.The single factor results with the greatest influence on the extraction effect were selected,and the response surface method was used to optimize the optimal extraction conditions at the level of 3 factors and 3 levels.The results were as follows:the ratio of material to liquid was 1:45,the extraction temperature was90?,the extraction time was 2 h,and the polysaccharide yield was 1.94%.The contents of total sugars,total phenols,total flavones,total triterpenoids and water,fat and protein in the extracts were detected respectively.The experimental results were as follows:total sugars 1.94%,total phenols 5.64%,total flavones 3.62%,total triterpenoids 2.46%,water 12.34%,fat 7.80%and protein 9.21%.The antioxidant activity of the extract of foraminifera obliquus was determined by investigating the scavenging ability of DPPH free radical,Fe2+ cod force and Fe3+reducing force of water extract.The experimental results showed that at the concentration of 5mg/mL,the clearance rate of DPPH water extract was 77.9%,which reached 79.3%of the clearance capacity of Vc in the control group.The net force of Fe2+ was 54.7%,which was 55.4%of the clearance force of EDTA in the control group.The content of Fe3+ was 0.65mm,up to 65.6%of the content of Vc in the control group.The above results indicated that the water extract of foramonas obliquus had good antioxidant capacity and showed dose dependenceEstablish a model of hyperlipidemia and hyperglycemia by zebrafish,and explore the hypoglycemic and lipid lowering ability of foraminifera betula.After soaking zebrafish with 111mM glucose solution for 15 days,the blood glucose value(14.65±0.43mmol/L)of zebrafish in the hyperglycemic group was about 8 times that of zebrafish in the control group(1.81 ± 0.14mmol/L).After successful modeling,they were immersed in the water extract of foraminifera obenzae with concentration of 1mg/L,10mg/L,50mg/L,100mg/L and 150mg/L for 7 days respectively.The results showed that the blood glucose value of the water extract group with concentration of 150mg/L was 2.94mmol/L,and the fructose amine level was 9.72mmol/L,which reached 65.5%and 94.4%of the hypoglycemic effect of the metformin group,showing a good hypoglycemic effect.In the hyperlipidemia experiment,only 10%yolk water was fed to the experimental group,and the contents of SOD,MDA,TG and TC in zeb fish were detected respectively.The experimental results were as follows:the SOD activity of the 150mg/L group was 19.87U/mg,and the SOD activity of the simvastatin treatment group was 15.16U/mg.MDA activity was 1.25mg/L in the 150mg/L concentration group and 1.02mg/L in the drug group.TG activity was 10.86mmol/L in the 150mg/L concentration group,9.34mol/L in the drug group,7.13mmol/L in the 150mg/L concentration group,and 4.12mmol/L in the drug group Showed good effect of lowering blood lipid.
