Medicinal fungi are one of the important sources of lead compounds,which can produce many novel skeletons and biological active secondary metabolites. Medicinal fungi have become a hot for scientific research workers to find and explore new drugs.As commonly used medicinal fungi, Coriolus versicolor and Lenzites betulina (L) Fr. both have better anti-inflammatory activity and have been the focus of active substances for the researchers. A variety of active ingredients in Coriolus versicolor, such as triterpenes, can anti-inflammation, inhibit the hepatitis B virus, regulate immune function, and inhibit platelet aggregation.Lenzites betulina (L.) Fr. also have anti-inflammatory activity, which was clinically used for waist pain, limb tic disorder, etc. Since there is less literature on these two medicinal fungi, it is important to research their value.With the development of society, there are an increasing number of inflammatory diseases, but the present anti-inflammatory drugs have a large and low security. To find safe and effective anti-inflammatory drugs is still an urgent problem.The thesis here focuses on the fungi identification, pharmacological activity and secondary metabolites from Coriolus versicolor and Lenzites betulina, guided by the anti-inflammatory activity evaluation. There are three chapters in this thesis.The first chapter is mainly about identification of Coriolus versicolor and Lenzites betulina by DNA molecular technique.In the second chapter, the anti-inflammatory activity of Coriolus versicolor was carried out in vitro. The results showed that Coriolus versicolor has no cytotoxicity to inflammatory cell; Coriolus versicolor showed strong anti-inflammatory activity on mononuclear macrophage (RAW)264.7 induced by LPS, with an inhibition rate of 28.88%. There experimental data confirmed its heat-clearing and anti-inflammatory effects recorded by the traditional literatures.The third chapter mainly about separation and purification through the normal phase silica gel chromatography, reversed-phase column chromatography, gel chromatography, HPLC chromatographic technology.Macrolactin A, Macrolactin E (1, 2) and 5, 7 - Octadecadienoic acid, 2, 3 -dihydroxypropyl ester were isolated from the 95% ethanol extracts of rice culture of Lenzites betulina. Macrolactin A and Macrolactin E are two large ring lactone class compounds and 5, 7 - Octadecadienoic acid, 2, 3 -dihydroxypropyl ester (3) is afatty acids compound. The three compounds were the first report to the best of our knowledge.In conclusion, this thesis reported the source identification,pharmacological-activity and secondary metabolites of Coriolus versicolor and Lenzites betulina, and the efficacy material foundation of the two medicinal fungi have also been carried out. The next research plan will be the anti-inflammatory and active metabolites of Lenzites betulina sporophore and the fermentation products of Coriolus versicolor.
