Coriolus versicolor(L.ex Fr.)Quel,also known as variegated Coriolus versicolor,C.versicolor,Bacillus,is a dry fruiting body of the genus Polyporaceae.CVFB flesh white,spore-colorless,cylindrical,mainly grown on a variety of broad-leaved tree piles,fallen wood and branches,especially grown on broad-leaved trees decay wood in summer and autumn,found all over the world,is the One of most medicinal value fungi.Diabetes is one of the most important chronic non-communicable diseases in the world,the number of adult patients with diabetes in the world is increasing year by year.However,the number of adult patients with diabetes in China ranks first in the world.The current research has not found any drugs or treatments that can cure diabetes,controlling and treating diabetes is a long-term,continuous and slow task,but,most of the diabetes drugs have toxic side effects while treating hyperglycemia symptoms.Long-term consumption of hypoglycemic drugs will increase the liver and kidney burden of the body and cause a series of complications.The natural drugs are natural products with certain pharmacological activities,including plants,animals and minerals et al.Natural medicines are easy to be absorbed and utilized by the human body,have a stable effect,and have little toxic and side effects on the body,natural medicine for diabetes can reduce the side effects of chemical drugs.Coriolus versicolor is a natural drugs has many pharmacological effects such as anti-tumor,anti-atherosclerosis,anti-aging,anti-oxidation and enhanced immune activity.At present,the research on Coriolus versicolor is mainly focused on macromolecular compounds such as polysaccharide,glycopeptide and glycoprotein,but Coriolus versicolor 's chemical composition is very complex.Therefore,this study determinated the main components of the CVFB from Heilongjiang province,and secondly,whether the CVFB has a therapeutic effect on ICR diabetic mice caused by streptozotocin(STZ),to provide a scientific basis for the development of drugs for the prevention and treatment of diabetes,it is of great significance to the in-depth study of Coriolus versicolor.In this paper,the components of CVFB from Heilongjiang origin were determinated by UV spectrophotometer,phenolsulfuric acid method,Kay-style nitrogen determination method,digestion method,Soxhlet extraction method,ignition quantitative method,inductively coupled plasma spectrum/ Mass spectrometry,high performance liquid chromatography and gas chromatography-mass spectrometry to detect total alkaloids,total flavonoids,reducing sugars,total triterpenes,total sugar,crude protein,crude fiber,crude fat,Total ash,metal content,mannitol,amino acid composition and fatty acid composition.STZ was used to construct a diabetes model for ICR mice.The mice were adaptively bred for 2 weeks.After fasting for 18 h,the STZ solution was administered intraperitoneally at a dose of 150 mg/kg.The mice were fed 2 h after the administration,after 7 days of feeding,mice were fasted(not fasted water)to measure fasting blood glucose,and ICR mice with fasting blood glucose >16.7 mmol/l were selected for the next experiment;The dose of CVFB powder was 1000 mg/kg,2000 mg/kg and 4000 mg/kg for 28 consecutive days by gavage,and the relevant indicators were determined after the end of the treatment period.The results of the study showed that CVFB can control or reduce the fasting blood glucose of mice and accelerate the rate of glucose metabolism;CVFB can reduce the levels of T-CHO,TG and HDL-C in mice,and regulate the abnormal symptoms of blood lipid metabolism commonly found in diabetes.It can increase the activity of SOD and GSH-PX antioxidant enzymes,enhance the ability of anti-oxidative stress in diabetic mice,reduce the levels of inflammatory factors TNF-? and IL-6 in the body,and reduce the damage of diabetes to liver;In the H&E staining and apoptosis experiments of pancreatic tissue,CVFB can greatly reduce the inflammatory factors present in islets,increase the islet cells,and reduce the apoptotic rate caused by diabetes.To provide a scientific basis for the development of drugs for the prevention and treatment of diabetes,it is of great significance to the in-depth study of Coriolus versicolor.
