Objective: To investigate the expression of cysteine aspartate-specific protease-14(Caspase-14,CASP-14),epidermal growth factor receptor(EGFR)and Bcl-2 in lesions of psoriasis vulgaris.To provide some reliable bases for exploring the etiology and pathogenesis of psoriasis and the clinical treatment of psoriasis vulgaris.Methods: A total of 60 samples were randomly selected from patients with psoriasis vulgaris diagnosed by pathologic biopsy from December 2013 to December 2014.20 samples of normal skin tissue were selected as controls.Hematoxylin-eosin(HE)was used to stain and observe the difference between non-keratinocytes and keratinocytes in normal skin and psoriasis vulgaris.The expression of Casepase-14,EGFR and Bcl-2 in normal skin tissue and psoriasis vulgaris lesions were detected by immunohistochemical method(Streptavidin-Peroxidase Immunohisto chemistry,SP-IHC)The results of gray staining were analyzed by image analysis software IPP6.0,and analyzed by statistical analysis software SPSS17.0.Results: Cysteine aspartate specific protease(Caspase-14)is involved in the construction of skin barrier one of the important factors.The skin is damaged by various harmful factors,the epidermal cells are damaged,the barrier function of the skin is destroyed,the skin of Caspase-14 will rapidly to the polyphenols(Pro-filaggrin)dephosphorylation and conversion into silk protein(Filaggrin),silk protein and then further hydrolyzed into a variety of natural moisturizing factor,to maintain moisture in the stratum corneum,involved in skin hydration,maintenance of the skin barrier function.In this study,the expression of Caspase-14 in psoriasis vulgaris lesions was significantly reduced(P <0.01),suggesting that this protein involved in the pathological changes of psoriasis vulgaris,directly or indirectly affect the skin barrier function.The expression of Caspase-14 in basal cell layer of psoriasis vulgaris was attenuated,and the expression of Caspase-14 was lower than that of normal control group The expression of cytoplasm in the cell layer of the spine cell layer and the granule layer was significantly lower than that in the normal control group(P <0.01).The expression of EGFR in the basal cell layer and spine cell layer of psoriasis vulgaris was significantly positive and positive,and there was significant difference between the two groups(P <0.01).The expression of Bcl-2 was significantly higher than that of the normal control group(P <0.01).The expression of Bcl-2 in the basal cell layer and the spine cell layer of the psoriasis vulgaris was significantly higher than that in the normal control group(P <0.01).Conclusion: 1.The expression of Caspase-14 in basal cell layer of psoriasis vulgaris was attenuated,and the expression of Caspase-14 was lower than that of normal control group The expression of cytoplasm in the cell layer of the spine cell layer and the granule layer was significantly lower than that in the normal control group(P <0.01).The expression of EGFR in the basal cell layer and spine cell layer of psoriasis vulgaris was significantly positive and positive,and there was significant difference between the two groups(P <0.01).2.Bcl-2 is a protective gene that inhibits apoptosis.It expresses Bcl-2 protein.Psoriasis lesions in both the excessive proliferation of keratinocytes,there are keratinocytes abnormal apoptosis.This study shows that the expression of Bcl-2 in basal cells is significantly reduced in psoriasis lesions,suggesting that the expression of Bcl-2 in basal cells is associated with an increase in apoptosis in psoriasis lesions.3.In normal epidermal tissue,EGFR mainly exists in the basal cell layer,and epidermal growth factor(EGF)and its family members through the tyrosine kinase pathway combined to promote the keratinocyte(Keratinocyte,KC)proliferation and differentiation.This study shows that EGFR expression in psoriasis vulgaris lesions was significantly increased,in the basal cell layer and the spine cell layer were significantly expressed,especially in the absence of mitotic granular layer expression,EGFR expression may be abnormal Leading to psoriasis vulgaris in the skin lesions of keratinocyte differentiation abnormalities,hyperplasia is one of the important reasons.4.The results showed that EGFR,Bcl-2 and Caspase-14 were closely related to the occurrence and development of psoriasis vulgaris,which was consistent with the current mainstream research.It not only provided for the improvement of psoriasis etiology and pathogenesis realistic evidence,but also for the clinical treatment of psoriasis and observe the efficacy of the objective basis.
