Flos chrysanthemum indicium has the effect of clearing heat,detoxification,clearing liver and improving vision.We found and refined a dual active site for the first time,with anti-hepatitis B virus and liver protection,which was made of chinese patent medicine-hepatoprotective pellet,quality standards were studied.To improve the quality standard of Flos Chrysanthemum indicum from diffrent origins by analyzing on linarin and cumambrin A qualitatively and quantitatively.In addition,we studied the extraction and separation technolgy of Flos chrysanthemum indicium yellow pigment,and effectively used the resources of Chinese medicine.Quality standard of Flos chrysanthemum indicium hepatoprotective pellet,we selected linarin,acacetin,cumambrin A and handelin for the index components.Qualitative analysis of the four indicator components was carried out by thin layer chromatography,TLC had obvious characteristics.The contents of the four indicator components were determined by HPLC with double wavelength(203 nm,340 nm).The content of linarin,acacetin,cumambrin A and handelin was 0.013%,0.063%,0.44%,0.50% respectively.The fingerprints of the pellet were studied by HPLC,14 common peaks were identified and seven compounds were identified,and the similarity was good.Solvent residues and pesticide residues were detected by Gas chromatographic,the residual residues of ethanol and ethyl acetate were 0.04% and 0.02%,respectively.The pesticide residues of ?-BHC and p,p'-DDT were 0.0012 mg / kg,0.003 mg / kg,others were not detected.Solvent residue and Pesticide residues were in line with the Pharmacopoeia.The heavy metal detection and moisture determination were studied,no heavy metals were detected and the water content was 1.5%.The results were in accordance with the Pharmacopoeia.The quality of the hepatoprotective pellets was effectively controlled by the experiments of thin layer identification,content determination,fingerprint,solvent residue and so on.Finally,we formulated the draft quality standard for hepatoprotective pellets.The quality control of Chinese Pharmacopoeia about Flos chrysanthemum indicium only determinated the content of linarin.We found that the sesquiterpenoids played a major antiviral effect,and the concent of cumambrin A varied with the quality of Flos chrysanthemum indicium.Therefore,we though the quality control of Flos chrysanthemum indicium should increase the sesquiterpenoids as index component.We used flavonoid and sesquiterpenoid as the index components.The qualitative analysis of linarin and cumambrin A was carried out by thin layer chromatography(TLC),TLC had obvious characteristics.HPLC method was used to determine the content of linarin and cumambrin A.The content of linarin in Wuyang,Jiyuan,Xinyang,Shandong and Anhui was 6.24%,5.02%,6.53%,4.79% and 6.19% respectively.The content of cumambrin A was 0.66%,0.75%,0.81%,0.47%,0.61%.Flos Chrysanthemum indicum from diffrent origins both contained linarin and cumambrin A,the content was different.It is the first time to establish a method to evaluate different components in Flos Chrysanthemum indicum by TLC and HPLC.The method is simple,accurate and reproducible,which can effectively improve the existing quality standard of Flos Chrysanthemum indicum.The result also showed Linarin and cumambrin A could reflect the quality of Flos Chrysanthemum indicum.The study on extraction and separation of yellow pigment in Flos Chrysanthemum indicum,we used the method of tissue fragmentation to study the extraction process of yellow pigment,and precipitation method was used to analyze the yellow pigment separation process.The extraction time was 3 times,the ratio of material to liquid was 1:10,the concentration ratio was 1/2,the concentration of water was 10%.In the process of magnification,the total yellow pigment was 60.19 g,and the yield was 1.2%.The results showed that the process was reliable and stable.In this paper,the draft quality standard of hepatoprotective pellet was established,which provided scientific basis for the quality control of traditional Chinese medicine.To improve the exisiting quality standards of Flos Chrysanthemum indicum,we increased cumambrin A as an indicator component.The extraction and separation of yellow pigment was studied to achieve the maximum utilization of Flos Chrysanthemum indicum resources.
