Study On The Improvement Of Quality Standard Of Erigeron Breviscapus(Vant.) Hand-Mazz.

Objective: To establish and improve the quality standards of Erigeron breviscapus,to provide a basis for controlling the quality of Erigeron breviscapus and establishing internal control quality standards.Methods:1.The characteristics and the Thin-layer chromatography(TLC)identification of Erigeron breviscapus to identify its authenticity.Character identification describes the morphological characteristics of the original plant and medicinal material;Identification of chlorogenic acid,scutellarin,3,5-dicaffeoylquinic acid,and 4,5-dicaffeoylquinic acid in breviscapus medicinal materials at different growth stages by thin-layer chromatography;2.The HPLC method was used to establish the fingerprints of Erigeron breviscapus medicinal materials,and the chlorogenic acid,baicalin,Contents of the four index components of 3,5-dicaffeoylquinic acid and 4,5-dicaffeoylquinic acid.3.The moisture,total ash,acid-insoluble ash,and extractives of Erigeron breviscapus in different growth stages were examined according to the four general methods of the Chinese Pharmacopoeia(2015 edition).Result:1.The characteristics of Erigeron breviscapus are obvious.The thin layer identification is good specific,which developed with methanol-water-formic acid(2: 2: 1)as the developing agent.When viewed at 365 nm,the spots are clear and the separation effect is good;2.A common pattern of fingerprints of breviscapus medicinal materials was established.Twelve batches of breviscapus medicinal materials fingerprinted 20 common peaks and identified 4 chemical constituents.The similarities between the fingerprints of the twelve batches of samples and the control fingerprints were all greater than 0.98.The HPLC conditions of Erigeron breviscapus were determined.Column: Agilent ZORBAX SB-C18(4.6 mm×250 mm,5 ?m);mobile phase: acetonitrile-0.1% phosphoric acid,gradient elution;flow rate: 1.0 m L·min-1;detection wavelength: 335nm;column temperature: 35?.3.According to the general rules of the Chinese Pharmacopoeia 2015 edition,the moisture content,total ash content,acid insoluble ash content and extract of the medicinal materials of Erigeron breviscapus were determined,and the results were all in accordance with the relevant regulations.Conclusion:1.Established an effective and controllable quality standard for internal control of Erigeron breviscapus.2.The high-performance liquid chromatography showed good specificity for fingerprints of Erigeron breviscapus.3.High performance liquid chromatography which has high precision,solution stability,accuracy and reproducibility can be used for the determination of the four index constituents in Erigeron breviscapus simultaneously.
