Objective: The periosteum distraction osteogenesis(PDO)combined with platelet-rich fibrin(PRF)was used to amplify the mandibular alveolar ridge of the rabbit,and the new bone formation was evaluated by using the patented distractor.Materials and Methods: 36 adult New Zealand white rabbits were randomly divided into three groups: A,B and C,12 rats in each group.All the experimental animals were removed from the left mandibular first premolars,1 month after the experiment.Group A(PDO group): left stretch area placed distractor.Group B(PDO + PRF): surgical method with A group,in the traction titanium mesh surface placed PRF membrane.Group C(xenotransplantation group): 5 * 5 * 5mm xenogeneic bone grafts on the left extraction zone,and covered with biofilm.The rats in group A and group B were sacrificed at 7 days,and the animals were sacrificed at the time of 0.25 mm / time,2 times / day,9 days for continuous operation,60 days for 120 days and 120 days for each group.Day were killed six animals.The observation of new bone formation: gross observation,new bone height measurement,X-ray analysis,Micro-CT examination,non-decalcified hard tissue section observation,HE histological observation,BMP-2 immunohistochemical observation.Results: PDO group fixed for 60 days,the traction area titanium mesh and traction around the visible part of the new bone formation,the average height of the new bone 2.65 ± 0.37mm;imaging shows that titanium under the network can be seen in the low density of new bone images,loose structure;Hard tissue sections and HE staining showed a large number of new trabecular bone,osteogenesis active.The bone height of the new bone was 4.13 ± 0.31 mm.The imaging showed that the new bone was thick and the trabecular bone was clear and the surrounding alveolar Bone integration as a whole;histological observation,new bone tends to mature,visible mature Harvard system,and autologous alveolar bone phase continued.Group B The above observations were similar to those in group A,and the bone grafts in group C showed a gradual absorption of the new bone.Conclusion: The rabbits were used to distract the rabbit mandibular alveolar bone membrane,fixed for 60 days,120 days found distraction gap with new bone formation.PRF did not show a significant effect on PDO.PDO is simple,small surgical trauma,no bone graft,has a great clinical value.
