The Pharmacodynamics Study Of The Wenweitiaochang Granula On Constipation-predominant Irritable Bowel Syndrome

Objective:To evaluate the protective effects of the Wenweitiaochang granula on the IBS-C symptoms and to explore its possible mechanism from the perspective of immune and gastrointestinal hormone,It will provide scientific basis for the development and application of the preparation.Methods:(1)After continuously intragastric administrating for 5 days,to observe the effect of WenweiTiaochang Granula on normal mice in intestinal first defecation time,defecation and number,quality,water content of the fecal.To observe the effect of Wenwei Tiaochang Granule on normal mice in gastric remnant rate and small intestinal propulsive rate by the intestinal propulsion and gastric emptying experiment.(2)Continuously intragastric administrated for 5 days,30 min after the last administration,mice in all groups except control group were given by introperitoneal injection of atropine sulfate,to observe the effect of WenweiTiaochang Granula on Atropine mice in intestinal first defecation time,defecation and number,quality,water content of the fecal.To observe the effect of Wenwei Tiaochang Granule on Atropine mice in gastric remnant rate and small intestinal propulsive rate by the intestinal propulsion and gastric emptying experiment.(3)To establish constipation mice model through thyroxine by gavage and water deprivation.To observe the effect of WenweiTiaochang Granula on Constipated mice in intestinal first defecation time,defecation and number,quality,water content of the fecal and the intestinal propulsion.(4)To observe the effect of Wenwei Tiaochang Granule on the pain response time,writhing latency and writhing number by Analgesic experiments in normal mice.(5)To establish constipation rat model by intragastric administration of compound diphenoxylate.To observe the effect of WenweiTiaochang Granula on constipation rat in intestinal first defecation time,defecation and number,quality,water content of the fecal.(6)To establish the rat model of constipation type of IBS by lavage for 0-4? ice water.After the treatment of Wenwei Tiaochang Granule,to take and weigh the thymus,spleen,to calculate viscera index,To detect the contents of MTL in the tissues of jejunum and plasma by elisa.To detect the ET and 5-HT levels in the plasma by elisa.To detect the CGRP.SP mRNA expression in the tissues of colon by RT-PCR.To detect the CGRP.SP protein expression in the tissues of colon by western bolt.Results:(1)The results of normal mice defecation experiment showed,Wenwei tiaochang Granule can shorten the time of the first defecation and increase the number and quality of the defecation.The results of mice bowel propulsion and gastric emptying experiments showed that It can significantly improve the rate of the small intestine and decrease the rate of gastric relict.(2)The results of Atropine mice experiments showed,Wenwei tiaochang Granule can shorten the time of the first defecation and increase the number and quality of the defecation.The results of Atropine mice bowel propulsion and gastric emptying experiments showed that It can significantly improve the rate of the small intestine and decrease the rate of gastric relict.(3)The results of the Thyroxine mice constipation model showed,Wenwei Tiaochang Granule can promote the weight gain of constipation mice,shorten the time of the first defecation,increase the number of defecation and increase the rate of small intestinal propulsion.(4)The results of the mice pain experiment showed,Wenwei Tiaochang Granule can prolong the mice's thermal pain response time,prolong the incubation period of the torsion and reduce the number of torsion.(5)The results of Diaphenoxylate rat constipation experiment showed,Wenwei tiaochang Granule can shorten the time of the first defecation and increase the number and quality of the defecation.(6)The results of model of IBS-C rat experiment showed,Wenwei Tiaochang Granule can improve the stool form and increase thymus and spleen index in rats,increase the content of plasma and jejunum tissues of MTL,decrease the content of ET and 5-HT in plasma,and decrease mRNA and protein expression level of CGRP and SP in colon.Conclusion:Wenwei Tiaochang granule have good treatment effect on constipation type of IBS-C,and improve symptoms significantly.Its mechanism may be related to increasing the content of MTL,reducing the content of ET and 5-HT and reducing CGRP and SP mRNA and protein expression in the tissues of colon.
