Serissa japonica(Thunb.)Thunb.,a kind of plant in Serissa genus of Rubiaceae family,is widely distributed in Jiangsu,Anhui,Zhejiang,Jiangxi,Guangdong,Yunnan and other regions of China.This paper is mainly for two aspects of S.japonica.First,the chemical composition of S.japonica was studied in order to find material basis of S.japonica.Second,anti-inflammatory on extract and monomer composition of S.japonica was researched so that we can find the pharmacological effects of its water extract,alcohol extract and monomer compounds.1 The chemical constituents of S.japonicaThe constituents were isolated by various chromatographic techniques(silica gel,RP-MPLC and PHPLC)from 70%ethanol extract and their structures were determined on the basis of their spectroscopic data,as well as literatures.Twenty compounds were obtained and identified as(?)-isolariciresinol(1),(+)-cycloolivil(2),(-)-vladinol D(3),(-)-(7'S,8S,8'R)-4,4'-dihydroxy-3,3',5,5'-tetramethoxy-7',9-epoxylignan-9'-ol-7-one(4),(-)-wikstrone(5),(+)-8-hydroxypinoresinol(6),(+)-8-hydroxymedioresinol(7),(+)-episyringaresinol(8),(+)-epipinoresinol(9),(-)-episyringaresinol(10),(-)-(7R,7'R,7"R,8S,8'S,8"S)-4',4"-dihydroxy-3,3',3",5-tetramethoxy-7,9':7',9-diepoxy-4,8"-oxy-8,8'-sesquineolignan-7",9",-diol(11),(-)-(7R,7'R,7"R,8S,8'S,8"5S)-4',4,"-dihydroxy-3,3',3",5,5'-pentamethoxy-7,9':7',9-diepo xy-4,8"-oxy-8,8'-sesquineolignan-7",9"-diol(12),8-hydroxyepipinoresinol(13),8-hydroxypinoresinol(14),8-hydroxymedioresinol(15),1,2-dimethoxy-3-hydroxy-6-methylanthraquinone(16),3-hydroxy-1,2-dimethoxyanthraquinone(17),1,2,4-trimerhoxy-3-hydroxy-6-methylanthraquinone(18),pinoresinol(19),medioresinol(20).Compound 10 is isolated from this plant for the first time,compounds 1?9 and 11?16 are isolated from Serissa genus for the first time.2 Anti-inflammatory research on extract and monomer composition of S.japonicaThe effect of aqueous extract,70%ethanol extract and major monomer components of different concentrations on RAW 264.7 cell viability was examined by MTT assay;The NO kit assay and the enzyme linked immunosorbent assay(ELISA)were adopted to detect the NO and interleukin-6(IL-6)release of extract and major monomer components on LPS-stimulated RAW 264.7 cells.We found that 70%ethanol extract of S.japonica and(7'S,8S,8'R)-4,4'-dihydroxy-3,3',5,5'-tetramethoxy-7',9-epoxylignan-9'-ol-7-one,(7R,7'R,7"R,8S,8'S,8"S)-4',4"-dihydroxy-3,3',3",5-tetramethoxy-7,9':7',9-diepoxy-4,8"-oxy-8,8'-sesquineolignan-7",9"-diol,episyringare sinol could inhibit the increase of NO and IL-6.
