Objectives: Many malignant tumor cells can secrete chemokines,which interact with tumor cells,study on chemokines has become one of the hot fields of cancer research;stromal cell derived factor is a class of small molecule protein chemotaxis,plays an important role in the differentiation and metastasis of tumor occurrence and development.Matrix metalloproteinases?MMPs?are a group with proteolytic function of zinc containing protein hydrolase family,are responsible for the degradation of extracellular matrix components in normaltissue remodeling and repair process,the excessive or inappropriate expression may be an important mechanism of invasion and metastasis of carcinoma.This article through the detection of stromal cell derived factor 1?SDF-1?and matrix metalloproteinase 9?MMP-9?expression in laryngeal squamous cell carcinoma and evaluate the roles of SDF-1 and MMP-9 expression in laryngeal squamous cell carcinoma and clinical significance.Methods: A retrospective study was conducted.42 cases paraffin-embedded specimens obtained from surgical resection with complete clinical data,and another 19 cases of adjacent laryngeal laryngeal mucosa were collected as controls from the Department of Otorhinolaryngology Head and Neck Surgery of the Neijiang First People's Hospital from February 2006 to February 2011.Survival and recurrence of laryngeal cancer patients were followed,as well as the clinical and pathological data.The immunohistochemical method?SP?was used to detect the expression of SDF-1 and MMP-9 in 42 cases of laryngeal carcinoma and 19 cases of adjacent laryngeal mucosa.The relation between the expression of SDF-1,MMP-9 and clinical pathological features and prognosis of patients with laryngeal squamous cell carcinoma were analyzed,as well as clinical data,such as age,sex,duration,pathology,TNM stage,clinical stage and follow-up results.Results:?1?The positive rate of SDF-1 in the laryngeal squamous cell carcinomas was 52.4%,which was higher than that in the adjacent negative laryngeal mucosa?21.1%??x2=5.250,P=0.022?.The positive rate of MMP-9 in the laryngeal squamous cell carcinomas was 59.5%,which was higher than that in the adjacent laryngeal mucosa?26.3%??x2= 5.772,P=0.016?;?2?There was no significant difference in the expression of SDF-1 and MMP-9 in laryngeal squamous cell carcinoma?LSCC?between different correlated,correlation index r=0.379,P=0.014;?9?The overall survival rate of SDF-1 positive expression patients was 45.0% and 81.8% of SDF-1 negative expression patients,the difference between them independent risk factor for the prognosis of patients,which was analyzed by COX risk regression model.Conclusions:?1?SDF-1 was highly expressed in laryngeal squamous cell carcinomas,but not related to pathological differentiation,suggesting that it was related to the tumorigenesis of laryngeal carcinoma.MMP-9 was highly expressed in laryngeal squamous cell carcinomas and had a higher expression rate in moderately and poorly differentiated tissues,suggesting that it was associated with the tumorigenesis and malignant differentiation of laryngeal squamous cell carcinoma;?2?The expression of SDF-1 and MMP-9 in laryngeal squamous cell carcinomas was not related to age,gender,duration of disease and smoking habit;?3?The expression of SDF-1 and MMP-9 in laryngeal squamous cell carcinomas was negatively correlated with T staging and clinical staging,suggesting that both of them may be related to the progress of laryngeal carcinoma;?4?The expression of SDF-1 and MMP-9 in laryngeal squamous cell carcinoma of patients with cervical lymph node metastasis was higher than that without metastasis,suggesting that both of them were associated with cervical lymph node metastasis;?5?The expression of SDF-1 and MMP-9 in laryngeal squamous cell carcinomas was not related to the clinical classification;?6?SDF-1 and MMP-9 in laryngeal squamous cell carcinoma were positively
