Objective:To investigate the effect of electroacupuncture at"Fenglong"on lipid metabolism in obese rats induced by high fat diet and the mechanism of related molecular network.Methods:Thirty-five male Sprague-Dawley rats were randomly divided into blank group(n=6)and high-fat group(n=29).The rats in the blank group were fed with normal diet for 16 weeks.The rats in the model group were fed with high fat diet for 12 weeks.The model of obese rats was established.The obese rat model was divided into model control group and electroacupuncture group according to the random number table.Electroacupuncture group of rats electro-acupuncture"Fenglong"points,each 30min,six days a week,treatment for four weeks.Blank group and model control group are synchronized and fixed.After 16 weeks to take tissue material,Observation:(1)Changes in body weight(2)Fat content around the kidney,epididymis,heart and the brown fat content after neck(3)The levels of plasma TC,TG,HDL and LDL were measured by biochemical method(4)HE staining was used to observe the histomorphology of liver in rats,including hepatic steatosis,inflammatory cell infiltration,edema,etc.(5)Tissue gene expression was changed and verified by qRT-PCR.Results:(1)Weight:the model group compared with the blank group,the weight was significantly increased,p<0.01;EA group and model group,compared with the 0th week to 2 weeks no significant difference,p>0.05,weight from the first 2 weeks Decreased,p<0.05;EA group 0 weeks and 4 weeks before and after weight comparison,weight decreased significantly,p<0.01.(2)Body fat:the model group compared with the blank group,the perirenal and epididyma l week fat was significantly higher,p<0.05,the peritumoral fat was significantly higher than the blank group,p<0.01,and the contrast after the brown fat Statistically significant,p>0.05;(3)Compared with the model group,the visceral fat mass(perirenal,epididymal week and heart weeks)was significantly lower than that of the model group(p<0.05),but the brown fat was significantly higher than that of the model group(p<0.05).(4)Serum index:Compared with the blank group,the content of TC and TG was significantly higher than that of the blank group,the content of HDL was significantly lower,the LDL had no statistically significant difference,p(TC)<0.01,p(TG)<0.05,p(HDL)<0.05,p(LDL)>0.05;Compared with model group,HDL and LDL were not statistically significant,p>0.05,while TC and TG contents were significantly lower than those in model group,p(TC)<0.01,p(TG)<0.05.(5)Microscopic observation:the size of the hepatocytes in the blank group was normal and homogeneous,the nucleus was in the middle of the liver cells,the hepatic sinusoids were arranged neatly,the cell membrane and cytoplasm were intact and the structure was clear and no obvious pathological changes were observed.Lesions,the organization appeared in a large number of different sizes of near-circular vacuoles,part of the nucleus was pushed to the side,part of the liver cell membrane and cytoplasm is not complete,accompanied by a large number of inflammatory cell infiltration;electroacupuncture group of liver cells extensive edema,Pulp loose,light stained,part of the cell membrane is not complete.The degree of hepatic cell fat lesion was reduced,the volume of vacuoles in the cells became smaller and the number of cells was decreased,and the degree of inflammation was also significantly reduced compared with the model group.(6)Results of gene chip detection:Compared with the blank group,992 genes were significantly downregulated in the model group and 1130 genes were significantly up-regulated.Compared with the model group,1092 genes were significantly upregulated in EA group and 1153 genes were significantly down-regulated.There were 496 genes,of which 246 were elevated in the model group and 250 in the model group at the same time.The results of path analysis showed that the above-mentioned common regulatory genes were mainly concentrated in Metabolic pathways,Neurotrophin signaling pathway,Salivary secretion,MAPK signaling pathway,Gap junction,Purine metabolism and so on.The expression of protein kinase C gamma,RPTOR,Ctnnbl1,Mapk11,Adrb1 and Prkaa2 were verified by PCR.The results showed that the above trends were consistent with the results of gene chip,the difference was statistically significant,p<0.05.Conclusions:High fat diet rats have increased body weight,fat configuration imbalance and elevated blood lipid status,accompanied by hepatocellular steatosis and inflammatory response.Electroacupuncture,"Fenglong"points through a variety of obesity-related gene targets,in the multi-channel formation of the network pathways,high-fat diet caused by lipid metabolism disorders have a positive regulatory role.
