Objective: Explore the action of endogenous thyrotrophin-releasing hormone (TRH) in the regulation of acupuncture analgesia and immunity and their possible conjunct mechanism.Methods: Inflammation and pain models in rat induced by Freund's Complete Adjuvant trigger as the research object, by using methods of behavioral threshold of pain test, high performance liquid chromatographic(HPLC) and radioimmunoassay et al ,we observed the influence of the content of 5-HT andβ-endorphin (β-EP) in hypothalamus or spinal cord, the level of IL-6 in blood serum, the changes of electro-acupuncture (EA) analgesia after acupuncture or injecting anti serum of TRH from the lateral ventricle of cerebrum on AA rats, to explore the relation of endogenous TRH with 5-HT orβ-EP on the effect of acupuncture analgesia of TRH in pain and acupuncture analgesia.Results: Electro-acupuncture can obviously increase the pain threshold of AA rats, improveβ-EP and 5-HT content in the hypothalamus and spinal cord, reduce IL-6 level. However, after injection of anti-serum of TRH from the lateral ventricle of cerebrum the amounts of 5-HT andβ-EP in hypothalamus of AA rats decreased, the IL-6 level raised,also the injection weakened the effect of EA.Conclusion: The electro-acupuncture has the dual role of analgesic and anti- inflammatory, the acupuncture analgesia and the regulation of the immune process most likely have a common mechanism, which can adjust the system by effecting their common cytokines, polypeptide and common receptors, formed a complete adjusting loop. Endogenous TRH is probably one of the common adjustment mechanisms in acupuncture analgesia and immune, may be related to the G-protein mediated signal transduction pathway.
