OBJECTIVE:To investigate the effects of Anshen Dingzhi Ling(ADL)on the concentration of glutamate and the expression of NMDAR1 and mGlu5 in the prefrontal cortex of the model rats for attention deficit hyper activity disorder(ADHD).METHODS:Fifty SHR rats were randomly divided into 5 groups:untreated model group,Methylphenidate group,low dose of ADL group,middle dose of ADL group and high dose of ADL group.The normal control group included 10 Wistar rats.The low,middle and high dose of ADL group were treated respectively with,the.crude dosage 6.7?13.4?26.7g/(kg·d)by intragastric administration 2 times a day;Methylphenidate group were treated with 2mg/(kg-d)Methylphenidate by intragastric administration 2 times a day;The untreated model and the normal control group were treated with double distilled water according to the body mass.The rats were killed after 4 weeks of treatment,then the concentration of glutamate and glutamine in the prefrontal cortex of rat brain were detected by ELISA while the protein expression of NMDAR1 and mGlu5 were detected by immunochemistry,and the mRNA transcription of NMDAR1 and mGlu5 in the prefrontal cortex of rat brain were detected by Real-time PCR.RESULTS:1.The concentration of glutamate and glutamine in prefrontal cortex in model group were significantly higher than normal group(P<0.01).after 4 weeks of treatment,the concentration of glutamate and glutamine in prefrontal cortex in low,middle,high dose of ADL groups,Methylphenidate group were significantly lower than the model group(P<0.01).2.The protein expression levels of NMDAR1 and mGlu5 in prefrontal cortex in model group were lower than normal group(P<0.05,P<0.01);after 4 weeks of treatment,the protein expression levels of NMDAR1 and mGlu5 were higher than the modle group(P<0.05,P<0.01)3.The mRNA transcription expression levels of NMDAR1 and mGlu5 in prefrontal cortex in model group were lower than normal group(P<0.05,P<0.01);after 4 weeks of treatment,the mRNA transcription expression levels of NMDAR1 and mGlu5 were higher than the modle group(P<0.05,P<0.01).CONCLUSION:1.ADL might improve ADHD symptoms by reducing the high concentration of glutamate and glutamine in prefrontal cortex of SHR rats.2.ADL might enhance the low protein and mRNA expression of NMDAR1 and mGlu5 in prefrontal cortex of SHR rats.3.ADL might cure ADHD by modulating the glutamate system.
