Coptidis Rhizoma is the dry stenophora of ranunculaceae plant Coptis Chinensis Franch,Coptis deltoidea C.Y.Cheng et Hsiao or Coptis teeta Wall.It can be digged in the autumn,the fibrous roots and silt should be removed,and then it should be dried and the remaining fibrous roots should be removed.For the different base resources,medicinal materials of coptidis rhizoma are divided into "Coptis chinensis Franch","Rhizaoma Coptidis from Yalin" and "Coptis teeta Wall".Coptidis Rhizoma is a kind of native medicinal material of Sichuan for Sichuan is the major production area of Coptidis Rhizoma.The Coptis chinensis Franch,Rhizaoma Coptidis from Yalin and the Sichuan frequently-used Coptis omeiensis(Chen)C.Y.Cheng(also called Pteris Coptis)are mainly produced in Sichuan.(1)The HPLC determination method was established firstly to test the alkaloid contents of seven coptis simultaneously in this study.The requirements of accuracy,reproducibility,accuracy,stability and linearity of the method were verified by methodology.In the study,the seven alkaloids contents and total alkaloids contents of 6 batches of Coptis chinensis Franch,6 batches of Rhizaoma Coptidis from Yalin,and 3 batches of Pteris Coptis were tested,and the content difference of total alkaloids and 7 alkaloids among the different species were compared,finally the chemical differences in three varieties was analyzed.The results showed that the total alkaloid contents of the Coptis chinensis Franch was the highest.However,the content of the seven alkaloids in three species was of significantly different.(2)Through the orthogonal experiment of four factors and three levels,the best processing technology of Jiuhuanglian,Jianghuanglian and Yuhuanglian was screened with the total alkaloid contents as the index.The total alkaloid contents of different processed products and raw products and the contents difference of seven kinds of alkaloids were compared in the study.The results showed that after processing,the total alkaloid contents of Yuhuanglian was the highest which was higher than that of raw products;the second one was curcuma which was also higher than that of raw products;and the total alkaloid content of Jiuhuanglian was lower than of that of raw products.To the respective contents of seven kinds of alkaloids,the contents were different for their own stable structures,and the change was also different after processed by different processing methods.(3)The HPLC specific chromatogram assay method of Rhizoma Coptidus was established in this study.The accuracy,reproducibility and stability of the method were verified in accordance with the requirements by methodology.The common characteristic peak 15 was found through the analysis of the similarity of the specific chromatogram of 15 batches of Coptis Chinensis.8 peaks were identified the alkaloid components of Coptis Chinensis by comparing with the reference substance of Coptis Chinensis.(4)In this study,the alloxan mouse diabetes model was applied to compare the pesticide effect differences of Coptis chinensis Franch,Rhizaoma Coptidis from Yalin and its processed products in the aspect of hypoglycemic.The results showed that the different Rhizoma Coptidis and it's different processed products have slight difference on the hypoglycemic effect,the Yuyalian has the most obvious effect on hypoglycemic;in the aspect of accommodation of glucose and lipid metabolism disorder and antioxidant,Rhizaoma Coptidis from Yalin has the best effect.
