Association Of VEGFC And VEGFA Gene Polymorphism With Diabetic Retinopathy In Uygur Population With Type 2 Diabetes Mellitus

Objective: Vascular endothelial growth factor(VEGF)is one of the most important factors responsible for Diabetic Retinopathy(DR).It has been reported that the variation of single-nucleotide polymorphisms(SNPs)of VEGFA and VEGFC genes are associated with DR in different ethnic groups.The present work was designed to investigate the relations between single nucleotide polymorphisms(SNPs)in the VEGFC and VEGFA gene and the development of DR in Uygur Population with type 2 diabetes mellitus(T2DM).Methods: Case-control study.A total of 302 patients with T2DM(164 participants with no history of DR [NDR],66 with non-proliferative DR [NPDR],72 with proliferative DR [PDR])and 333 Normal Controls(NC)were recruited.Three VEGFC(rs17697419,rs17697515,rs2333526)and five VEGFA(rs3025021,rs10434,rs833061,rs2010963,rs699947)SNPs were genotyped by PCR-RFLP system.Results: Multiple tag SNPs in the VEGFA and VEGFC gene were associated with PDR.The G/A genotype and the A alleles rs17697419(P=0.019,P=0.021,respectively),the C/C genotype and C alleles of rs2010963(P=0.017,P=0.039),the A/A genotype and the A alleles of rs699947(P=0.028,P=0.041),the T/T genotype and the T alleles of rs3025021(P=0.019,P=0.038),the C/C genotype and the C alleles of rs833061(P=0.028,P=0.021)were associated with PDR.A total of 11 haplotypes with frequency >1% were obtained of the 5 SNPs in VEGFA,3 haplotypes with frequency >1% were obtained of the 3 SNPs in VEGFC using Haploview software.Haplotypes ACG(38.3%),CTC(32.7%)and CTG(28.8)were the three most common haplotypes obtained of VEGFC.Conclusions: The polymorphisms of VEGFA and VEGFC might associated with PDR.
