Objective:To observe the effect of Wen Shen Gu Shu decoction on bone mineral density and the expression of inflammatory cytolines of ovariectomized rats with postmenopausal osteoporosis.Material and Method:Post menopausal osteoporosis(PMOP)rats model was established by excision bilateral ovaries of 9-month-old Sprague-Dawley female rats.After successful modeling,48 modeling rats were randomly divided into model group,estradiol valerate group,Wen Shen Gu Shu decoction groups with different doses.Rats with resection of abdominal adipose tissue were divided into sham group.The changes of bone mineral density(BMD),Interleukin-1 ?(IL-?),tumor necrosis factor-?(TNF-?),Interleukin-6(IL-6)and transforming growth factor-?(TGF-?)were detected respectively after 12 weeks.Result:1.Effect of Wen Shen Gu Shu decoction on BMD in rats with postmenopausal osteoporosis:after modeling,compared with model group and sham group,ovariectomized rats in the model group,the lumbar vertebra BMD decreased significantly(P=0.006<0.01),suggesting that after modeling bone mineral content and serious loss;after drug treatment,compared with model group,Wen Shen Gu Shu decoction with medium dose group rats BMD increased significantly(P=0.03<0.05),and the high dose group BMD increased more significantly(P=0.006<0.01),they both had statistical difference(P<0.05).Except for the low dose group(P=0.033<0.05),there was no statistical difference between the high and medium dose group and estrogen group(P>0.05).2.Effect of Wen Shen Gu Shu decoction on the changes of IL-1??TNF-? and IL-6 in rats'Proinflammatory cytokines of postmenopausal osteoporosis:after modeling,compared with model group and sham group,ovariectomized rats in the model group,the peripheral serum Proinflammatory cytokines IL-1?,TNF-? and IL-6 were significantly increased in ovariectomized rats(P<0.01,P values were 0.006,0.001 and 0.003 respectively).After drug treatment,compared with model group,Wen Shen Gu Shu decoction with high dose group rats serum IL-1? and TNF-?and IL-6 content were significantly decreased(P<0.01,P values were 0.003,0.007 and 0.006 respectively);middle dose group rats serum IL-1? and TNF-? and IL-6 content decreased significantly(P<0.05,P values were 0.004,0.023 and 0.014 respectively);low dose group rats serum IL-1? and IL-6 content decreased significantly(P<0.05,P values were 0.009 and 0.027 respectively),although TNF-? content was also decreased,just no statistical differences(P=0.065>0.05).There was no statistical difference between the groups in the treatment of Wen Shen Gu Shu decoction and the estrogen group(P>0.05).3.Effect of Wen Shen Gu Shu decoction on the expression of TGF-P mRNA in rats' bone tissue of postmenopausal osteoporosis:after modeling,compared with model group and sham group,ovariectomized rats in the model group,the expression of TGF-? mRNA in bone tissue of ovariectomized rats was decreased(P=0.037<0.05).After drug treatment,compared with model group,the expression of TGF-? mRNA in Wen Shen Gu Shu decoction with high dose group rats'bone tissue was increased significantly(P=0.016<0.05)and the rest of the intervention group expression although there is an increase,but no statistical difference(P>0.05).Conclusion:1.Wen Shen Gu Shu decoction can effectively prevent and treat PMOP.2.Wen Shen Gu Shu decoction in the treatment of osteoporosis mechanism may be related to the down-regulation of IL-1?,TNF-?,IL-6 of proinflammatory cytokines expression,inhibit bone resorption;while up regulation of TGF-?,inhibiting the expression of anti-inflammatory cytokines,promote bone formation.
