ObjectiveIn this paper,the traditional Chinese herbal medicine Salvia miltiorrhizae as the research object from non-GAP base.Residue level of pesticide and aflatoxin in medicinal herbs was respectively determined by gas chromatography and liquid chromatography-mass spectrometry,and the antimicrobial effect of medicinal materials was investigated by microcalorimetry.Combined with chemometrics to evaluate the antibacterial effect of multi-batch Salvia miltiorrhizae,the main antimicrobial activity was explored by establishing the "spectral-effect" relationship between chemical fingerprints and antibacterial activity of medicinal materials,in order to improve the safety and antibacterial activity of Salvia miltiorrhizae evaluation to provide reference and basis.Methods1.Pesticide residues in Salvia miltiorrhizae were determined by the optimized pesticide residue method accoding to Ch P.(2015 edition)and the results were confirmed by UFLC-MS/MS.2.Quantification and qualitation of aflatoxins(B1,B2,G1,G2)in Salvia miltiorrhizae were performed by LC-MS/MS after immunoaffinity column clean-up.3.The effects of Salvia miltiorrhizae on anti-staphylococcus aureus,anti-resistant staphylococcus aureus and anti-escherichia coli were studied by microcalorimetry.4.Chemical fingerprint and biothermal activity based on HPLC and microcalorimetry were combined for exploring the antibacterial components of Salvia miltiorrhizae.Results1.The residues of organochlorines,organophosphorus and pyrethroid pesticides from 32 batches of Salvia miltiorrhizae were determined that the methodological validation(including instrument precision,sample recovery rate and stability)was in accordance with the regulations.The results showed that seven kinds of organophosphorus and pyrethroid pesticide residues were detected.A total of 10 batches were detected cypermethrin,and the highest content of monocrotophos was 0.028 mg/kg.The above results were verified by UFLC-MS / MS method,avoiding false positive results.2.The results showed that the optimized method was more reasonable and reliable than the traditional method.Through MRM scanning and EPI retrieval,it can achieve simultaneously qualitative and quantitative analysis of multiple-mycotoxins residues.3.The antimicrobial effects of Salvia miltiorrhizae on Staphylococcus aureus,resistant Staphylococcus aureus and Escherichia coli were evaluated by microcalorimetry.The results showed that it was various antibacterial activities in different origin of Salvia miltiorrhizae,such as purchased from Beijing in five producing origins the highest resistance to escherichia coli,On the contrary,the weakest against staphylococcus aureus,indicating that the antibacterial activity of Salvia miltiorrhizae was interaction of various components.4.Combined with cluster analysis,principal component analysis and multiple linear regression analysis,which were explored the chemical fingerprint of Salvia miltiorrhizae and its effect on pseudomonas aeruginosa.The results showed that the inhibition rate(?,%)of samples collected in Beijing was the highest.On the contrary,the?of samples collected in Sichuan province was the lowest,which indicated that the anti-pseudomonas aeruginosa was more resistant to lipophilic components.The results showed that peak 4,peak 8,peak13,peak 17 and peak 18 in the fingerprint were the main five antimicrobial active substances.ConclusionsIn this paper,investigate the residual level of aflatoxin and pesticide in Salvia miltiorrhizae,and its effect on a variety of bacteria were determined by the establishment of the "spectral-effect" relationship between the chemical fingerprint and antibacterial activity.The safety and antimicrobial differences of varieties origins of Salvia miltiorrhizae were evaluated,in the clinical practice is more suitable for which to provide a theoretical basis for its clinical rational and safe application.
