OBJECTIVEIn the 19th century, the hypothesis, tumor originated from chronic inflammation,was first proposed by Rudolf Virchow. Since then, the inflammatory reaction is known as one of the seventh largest biological characteristics of malignant tumor, which is closely related with gastric cancer occurrence, progression, metastasis,invasion, etc. The mechanism that tumor induced by inflammation has aroused people's enduring research .However,the environment inflammatory cytokines IL - 9 and mast cell (MC) are becoming more and more popular .Experiments in this paper are going to observe the distribution of IL-9 and MC in chronic non-atrophic gastritis, gastric precancerous lesion tissues (chronic atrophic gastritis with intestinal, intraepithelial neoplasia) and gastric cancerous tissues, and to explore its' clinical significance, which will help speculate the effect of IL-9 and MC in the development of gastric cancer induced by chronic inflammation and provide beneficial theoretical basis in the control and prevention of gastric precancerous lesion tissues.METHODSRetrospective study method was used in this research. The specimens from gastroscope biopsy and stomach surgery were selected from inpatients of Binzhou people's hospital in July 2013 to July 2014, including chronic non-atrophic gastritis,gastric precancerous lesion tissues (chronic atrophic gastritis with intestinal,intraepithelial neoplasia) and gastric cancerous tissues. All samples must be approved by the following process: part of the sections used HE stain to observe the stomach tissue morphology; part of the sections used W-S silver stain to observe the presence of H.pylori infection, part of the sections used immunohistochemical method to observe IL-9, MC distribution. At last, to analyse them with clinical pathological factors.RESULTS1.1.1 IL-9 was mostly expressed in the cytoplasm and nuclear of lymphoid cells.The positive rates in chronic non-atrophic gastritis, gastric precancerous lesion tissues and the gastric cancer group were respectively 8.33%, 30.00% and 4.35%,the differences of which were statistically significant (P=0.035).1.2 In gastric precancerous lesion tissues, the expression of IL-9 in patients with H.pylori (+) was significantly higher than that of patients with H.pylori (-), while the higher degree of inflammation, atrophy and intestinal metaplasia, the expressin of IL-9 were increased, and differences were statistically significant. (P values are: 0.013, 0.001,0.023, 0.001; r values are: 0.451, 0.557, 0.448, 0.543);but age, sex, activity had no relationship with the expression of IL-9.22.1 MC were mainly distributed in mucosa and submucosa. In chronic non-atrophic gastritis, the average number of MC in gastric benign lesion tissues,gastric precancerous lesion tissues and the gastric cancerous tissues were respectively 31.96±9.11.38.97±10.24.20.79±10.94,the differences of which were statistically significant (P=0 .000).2.2 In gastric cancerous tissues, the average number of MC with H.pylori (+) was higher than that of patients with H.pylori (-)(26.80±11.62?16.17±8.09),and the differences were statistically significant (P=0.017).In gastric precancerous lesion tissues also indicated the differences between H.pylori (+) and H.pylori(-) groups were statistically significant(P=0.001), besides the rest of the various indicators had are no correlation with the quantity of MC (P > 0.05).3 In gastric precancerous lesion tissues ,the MC average number in specimens with IL-9(+) was higher than that of IL-9(-) (45.850±2.742?37.878±2.188),and the difference is statistically significant (P = 0.045).CONCLUSIONIL-9 and MC are highly expressed in gastric precancerous lesion tissues, which are related with H.pylori infection. Moreover, the IL-9 expression was correlated with MC. All of those proved that IL-9 and MC participate in the process of chronic gastritis to cancer, especially in the stage of gastric precancerous lesion. But the specific mechanism remains to be further research.
