Objective:1)To detect the expression of Flotillin-2 protein in specimens of gastric cancer and adjacent tissues by immunohistochemical staining,and to investigate the relationship between their expression and clinicopathological features and prognosis of gastric cancer.2)To further explore the clinical value of Flotillin-2 protein in predicting lymph node metastasis and prognosis in human solid tumors by performing a synthetic analysis with relevant literatures.Methods:1)The expression level of 112 cases of gastric carcinoma and paired para-carcinoma tissues(from the edge of the cancerous tissue > 5 cm)was detected by immunohistochemical staining.The correlation between Flotillin-2 protein expression and clinicopathological characteristics and prognosis was statistically analyzed.2)An comprehensive literature retrieval was conducted to collect the relevant published articles that reporting the association between Flotillin-2 expression and lymph node metastasis or prognosis.Its clinical significance for predicting lymph node metastasis and prognosis in solid tumor was investigated.Results:1)Immunohistochemical data showed that the Flotillin-2 protein was mainly expressed in the cytoplasm of tumor cells,and basically no expression in paracarcinoma tissues.The expression level of Flotillin-2 in cancer tissues was significantly higher than that of adjacent tissues.There was no association between Flotillin-2 protein expression and gender,age,tumor location and differentiation(P greater than 0.05).However,its protein expression showed an significant association with tumor size(P=0.001),depth of invasion(P=0.005),lymph node metastasis(P <0.001),distant metastasis(P < 0.001)and AJCC stage(P < 0.001).Kaplan-Meier survival analysis revealed that in the Flotillin-2 low-expression group the overallsurvival rate was significantly higher than that of the high expression group,and the difference between two groups had a statistical significance(P < 0.001).Multivariate regression analysis showed that distant metastasis,AJCC stage and Flotillin-2 protein expression were independent prognostic factors of gastric cancer.2)The meta-analysis further confirmed the clinical significance of Flotillin-2protein in human solid tumors,indicating that its expression was closely related with lymph node metastasis and overall survival.The patients with high expression of Flotillin-2 in cancerous tissue were prone to lymph node metastasis and had a shorter overall survival time.Conclusion:The high expression of Flotillin-2 was closely related to the development and metastasis of gastric cancer.There was an association between Flotillin-2 expression and clinical-pathological features and prognosis in gastric cancer.And the meta-analysis further revealed that Flotillin-2 protein expression has important clinical significance in human cancer,and can be used as a valuable indicator for the evaluation of lymph node metastasis and prognosis of solid tumors.
