Objective:The aim of this study was to investigate the effect of daidzein and its derivatives on cell proliferation and apoptosis in non-small cell lung cancer A549,H1299 cells and lung normal cells HELF cells in order to explore the effect of daidzein and derivatives on non-small cells anti-tumor effect in lung cancer.Method:1.10-5,10-4,10-3,10-2,10-1,1,10,100,1000?M soybean isoflavones and derivatives were used in A549,H1299 and HELF cells for 6h.CCK-8 was used to investigate the effects of soybean isoflavones and derivatives on the proliferation of A549,H1299 and HELF cells.2.After 100?M of soybean isoflavones extract,10?M of daidzein,100?M of derivative A,10-1?M of derivative B,1?M of derivative C treatment on A549,H1299,HELF cells for 0,1,3,6,12 and 24 h,the effect of soybean isoflavones and derivatives on the proliferation of A549,H1299 and HELF cells was detected by CCK-8 method.3.After 100?M of soybean isoflavones extract,10?M of daidzein treatment on A549,H1299,HELF cells for 6h,the effect of soybean isoflavones on the apoptosis of A549,H1299 and HELF cells was analyzed by flow cytometry.Results:1.1000 ?M soybean isoflavone crude extract acted on A549 cells 6h,compared with DMSO control group,can significantly reduce the activity of A549 cells and inhibit their proliferation(P<0.05).Daidzein inhibited cell proliferation of H1299 cells in the range of 10 to 1000 ?M,and the inhibitory effect was time-dependent.The inhibition of proliferation occurred mainly after 12 h.The effect of ramification A on the proliferation of A549 cells in the range of 10-1 to 100 ?M was dose-dependent.The effect of ramification B on cell proliferation inhibition of A549 cells in the concentration range from 10-1 to 1000 ?M.In the 1 to 100 ?M concentration range,the ramification C inhibited cell proliferation of H1299 cells,and the inhibitory effect was time-dependent.The inhibition of proliferation occurred mainly after 6h.2.The effects of soybean isoflavones on the apoptosis of A549,H1299 and HELF cells were studied by flow cytometry.The results showed that the crude extracts of soybean isoflavones were treated with 100?M soybean isoflavone extract on A549,H1299 and HELF cells for 6 hours(P <0.05),and inhibited the apoptosis of HELF cells(P <0.05),and promoted the apoptosis of lung cancer cells A549 and H1299 cells significantly(P <0.05).Daidzein could significantly promote the apoptosis of A549 cells(P <0.05),and inhibit the apoptosis of HELF cells(P <0.05)after treatment with A549,H1299 and HELF cells for 6 hours with 10?M daidzein.The above results showed that the apoptosis of A549 and H1299 cells was induced by soybean isoflavones at the cellular level.Conclusion:Soybean isoflavones and derivatives can inhibit the proliferation of lung cancer A549 and H1299 cells,and the inhibitory effect is time-dependent and dose-dependent,and soybean isoflavones inhibit the proliferation of lung cancer A549 and H1299 cells may be related to the induction of cancer cell apoptosis.Soybean isoflavones and derivatives can promote the proliferation of HELF cells in a time-dependent manner,and soybean isoflavones can inhibit the apoptosis of HELF cells.
