ObjectiveIn the world,male infertility accounts for 40-50%of the total infertility population,which affects about 7%of all men.Among the many causes of male infertility,vitamin D is thought to be directly related to male fertility.The aim of this study was to explore the relationship between vitamin D and semen quality in males through by the overall levels and local levels of vitamin D which is serum vitamin D and seminal plasma vitamin D respectively.In order to study the specific mechanism of vitamin D on male reproductive mechanism to provide a more comprehensive research direction.In order to provide a more comprehensive research direction to study the specific mechanism of vitamin D on male reproductive.And study its specific pathways to sperm by incubate the spermatozoa with the activated form of vitamin D-1,25(OH)2D3 in vitro.MethodsPart I Clinical analysis between serum and seminal plasma vitamin D with sperm parametersAccording to the inclusion criteria and the exclusion criteria,the semen samples were collected from male patients and normal subjects who were admitted to the male medical clinic in our hospital.Application of WLJY-9000 Weili color sperm quality detection system to detect sperm concentration,sperm count,sperm motility,percentage progressive sperm motility and corresponding kinematic parameters including curve velocity(VCL),linear velocity(VSL)and average path velocity(VAP)]according to WHO "Human Semen Examination and Processing LaboratoryManual"(fifth edition).Acridine orange staining was used to detect sperm DNA fragmentation index(DFI)and high DNA dyeability(HDS).The sperm morphology was carefully evaluated by the WHO criteria using modified Papanicolaou staining.Serum and seminal plasma vitamin D levels were detected by Electrochemiluminescence method.And then analysis the relationship between serum and seminal plasma vitamin D with the parameters of semen.Part ? The effects of 1,25(OH)2D3 on sperm motility and mechanismAccording to the inclusion criteria and the exclusion criteria,the semen samples were collected from male patients and normal subjects who were admitted to the male medical clinic in our hospital.The percoll density gradient centrifugation was used to extract the spermatozoa.The sperm corresponding kinematic parameters were detected by WLJY-9000 Weili color sperm quality detection system after incubation with the 1,25(OH)2D3 at different concentrations(0,0.01nM,0.1nM,1nM,10nM)and different time(0,15min,30min,60min,90min).According to the preliminary results,the spermatozoa treated with optimal concentration of 1,25(OH)2D3 for 30 min were selected to detect the altitude of sperm ascending and the contents of cAMP,NADH and ATP and the activity of spermatozoa PKA.ResultThere was no relationship between the level of serum vitamin D and the parameters of sperm(p>0.05).Seminal plasma vitamin D positively correlated with sperm motility(p = 0.026),percentage progressive sperm motility(P = 0.031),VCL(p=0.005),VSL(P = 0.011)and VAP(P = 0.005).Sperm motility parameters increased the most significant after incubation with the 1,25(OH)2D3 which the concentration was 0.1 nM by 30 min.The sperm ascending altitude was remarkably increased.The spermatozoa cAMP,NADH and ATP contents were significantly increased(p<0.05),and also the spermatozoa PKA activity was significantly enhanced(p<0.05)on this condition.ConclusionSeminal plasma vitamin D may be involved in the regulation of sperm motility.The active form of vitamin D may enhance the sperm motility by promoting theaction of spermatozoa mitochondrial electronic respiration chain through cAMP/PKA and NADH pathways by increasing ATP production.
