Sambucus williamsii Hance is one of the plants of Sambucus linn belonging to Caprifoliaceae,it is deciduous shrub or dungarunga,which is also called Jie-gu-dan,Gong-dao-lao,Shu-jin-shu.Wild resources are abundant in China,distributed widely in the Northeast,North China and Inner Mongolia.It is a kind of traditional Chinese medicine with a long history of medicinal applications,sweet and bitter in taste,flat in nature and non-poisonous.Because of the efficacy of promoting blood circulation,alleviating pain,expelling the wind and promoting reunion of bone,it is usually used as a cure of bruises,swelling,pain,trauma hemorrhage and fractures.Clinical researches show that the extract of roots of Sambucus williamsii Hance is characterized by short treatment course,high curative effect and non-poisonous or side effects when used to treat fractures.Our research groups had confirmed the effective part of roots to promote fracture healing through pharmacodynamics,chemical constituents of the effective part were studied preliminarily.Experiments were done on the base of early work of our research groups,chemical constituents from roots of Sambucus williamsii Hance were studied systematically and deeply,to illustrate material basis of pharmacodynamics of the effective part,formulate the quality standards for medicines,medicinal materials and research on biological activity further.To enrich the effective part through AB-8 macroporous resin,chemical constituents of 50%ethanol elution fractions were isolated by morden methods,such as silica gel column chromatography,ODS column chromatography,preparative and semi-preparative HPLC,twenty-two compounds were obtained.Their chemical structures were elucidated on the basis of spectral data of 1D-NMR such as 1H-NMR,13C-NMR and NOE,2D-NMR such as 1H-1H COSY,DEPT,HSQC and HMBC,combined with spectral data of HR-ESI-MS.Fifteen compounds compounds were identified,including five iridoid glycosides,they were identified as ?-D-apiofuranosyl(1"?6')-8-epi-loganin,a-morroniside,sweroside,loganin and dehydromorroniside;five alkaloids,they were identified as 5-(1'-hydroxyethyl)-methyl nicotinate,3-(hydroxyacetyl)indole,3-methoxyl-1H-pyrrole,4'-hydroxy-N-(4-hydroxy-3-methoxybenzoyl)-3',5'-dimethoxy benzamide and(1S,3S)-1-methyl-1,2,3,4-tetrahydro-?-carboline-3-carboxylic acid;two lignans,they were identified as(7R,8R)-7,8-dihydroxyl-9'-hydroxyl-3'-methoxyl-8-hydroxylmethyl-7-(4-hydroxyl-3-methoxyphenyl)-1'-benzofuranpro panol-9'-O-?-D-glucopyranoside and(7R,8R)-3-methoxy-8,4'-oxyneolignan-3',4,7,9,9'-pentol(threo);one phenylpropanol,it was identified as coniferyl alcohol 9-O-?-D-glucopyranoside;one phenolic acid,it was identified as syringic acid-4-O-a-L-rhamnopyranoside;one sterol,it was identified as ?-sitosterol.Via network platform of SciFinder,?-D-apiofuranosyl(1"?6')-8-epi-loganin was a new compound,seven compounds were firstly isolated from Caprifoliaceae,two compounds were isolated from Sambucus L.for the first time.Studies had been systematically done on chemical constituents of effective part of promoting fracture healing.In order to investigate pharmacodynamics and mechanism of iridoid glycosides and lignans components,the number and kinds of monomers had been preparated,material basis was provided for exploitation and utilization.
