Sambucus williamsii Hance is one species of Sambucus genera in Caprifoliaceae.It has the potence of expelling wind-dampness,rejoining fracture and restoring soft tissue,invigorating blood circulation and stopping pains.Our previous study found that the 60%ethanol crude extract of the stems of Sambucus williamsii Hance was eluted by D101 macro adsorptive resin column with a gradient of ethanol/water(0,30%,50%and 95%),and JGM(the fractions of 50%and 95%)was considered as the antiosteoporosis active fraction by increasing the proliferation activities of UMR106 cells and preventing ovariectomy-induced bone loss in rats.To explore the potential bioactive constituents,we have a systematic chemical investigation on the antiosteoporosis active fraction of Sambucus willamsii Hance.29 compounds were isolated from JGM through various chromatographic techniques,including 15 lignans,8 phenolic acids,3 triterpenoids and 3 other kinds of compounds.The structures were elucidated on the basis of theirs physicochemical properties and spectroscopic characters as(7R,8S,7'R,8'S)-syringaresinol(1);(7R,8S,7'R,8'S)-medioresinol(2);(7S,8R,7'S,8'R)-pinoresinol(3);(7S,8S,7'R,8'S)-lirioresinol A(4);(7S,8R,7'S,8'R)-1-hydroxypinoresinol(5);(7R,8S)-ficusal(6);(7R,8S)-5-methoxybalanophonin(7);(7R,8S)-balanophonin(8);(7R,8S)-dihydrodehydrodiconiferyl alcohol(9);(7R,8S)-5-methoxy-trans-dihydrodehydrodiconiferyl alcohol(10);(7R,8S)-1-(4-hydroxy-3-methoxyphenyl)-2-[-4-(3-hydroxypropanyl)-2-methoxyphenoxy]-1,3-propanediol(11);(7S,8S)-1-(4-hydroxy-3-methoxyphenyl)-2-[-4-(3?hydroxypropanyl)-2-methoxyphenoxy]-1,3-propanediol(12);(7S,8S)-guaiacylglycerol-?-O-4'-conifery ether(13);(7S,8R,7'S)-cycloolivil(14);(7S,8R,8'S)-isolariciresinol(15);vanillin(16);p-hydroxybenzaldehyde(17);octadecyl-(E)-ferulate(18);eicosyl-(E)-ferulate(19);docosyl-(E)-ferulate(20);2-propenoic acid,3-(4-hydroxy-3-methoxyphenyl)-(9Z)-9-octadecenyl ester,(2E)-(9C1)(21);octadecyl-(E)-caffeate(22);eicosyl-(E)-caffeate(23);23-hydroxy-3-oxo-28-nor-12,16-dien oleanane;Sambucusan A(24*);oleanolic acid(25);3-oxooleanolic acid(26);octacosanol(27);hexadecanoic acid(28);?sitosterol(29).Among them,1 compound(24)was a new compound and 11 compounds(5,7,8,10,17-23)were isolated from the Sambucus genus for the first time.The proliferation effects of 28 isolated compounds on osteoblast-like UMR106 cells were determined.The results showed that 18 compounds(3?17,21,24,25)significantly increased UMR106 cell proliferation.Furthermore,the study has also established the HPLC specific chromatogram of the antiosteoporosis active fraction from Sambucus williamsii Hance,and identified the main peaks(occupied more than 60%total peak areas)by all isolated compounds.Our present investigation provided the scientific evidence for research and developement of Sambucus williamsii Hance and its related preparations on osteoporosis in the future.
