Analysis On The Chemical Constituents In Vitro And Study Of Constituents Absorbed Into Blood Of Sanjie-Zhentong Capsule

Sanjie-Zhentong Capsule is composed of four herbals including D.cochinchinensis,Panax Notoginseng,Fritillariae Thunbergii Bulbus,Coicis Semen,which can resolve the hard lumps,resolve the hard lumps,soften and resolve hard mass.Sanjie-Zhentong Capsule is used for endometriosis(EMs)and secondary dysmenorrhea,irregular menses,pelvic bag piece and infertility that are cuased by EMs.However,as Chinese medicine compound preparations,because of complex composition,the active constituents of the capsule are hard to clarify and the development of the capsule is restricted.The mian task of this topic is focused on the vitro and in vivo changes of chemical composition of Sanjie-Zhentong capsule,lay the foundation for the further study on the material basis of Sanjie-Zhentong capsule to the treatment of endometriosis.In the further,the result of this study can be used to guide the optimization of the technology and the improvement of the quality standard for the capsule.The study comprises two parts:The first part is the chemical analysis about Sanjir-Zhentong Capsule in vivo.The preliminary analysis of 45 chemical constitutents of Sanjie-Zhentong capsule have been made by UPLC/Q-TOF-MS technology.Unite the informations of the online spectrograms of UV,ESI-MS,ESI-MSMS to identify 38 chemical constitutents,among this constitutents,13 constitutents were identified by compared with the reference compounds.In the meantime,the feasible fragmentation pathways of Flavonoids,Saponins,and Alkaloids were summarized to emhance the accuracy for the identification of the compounds.And on the basis of preliminary research about the analysis and identification of the composition of the capsule,establish two HPLC-UV fringerprints of Sanjie-Zhentong capsule,for detect and control the preparation stability and qualitive consistency of Sanjie-Zhentong capsule,so as to provide stable quality Sanjie-Zhentong capsule for the research of the components in drug-containing plasma.The second part is the study of serum pharmacochemistry of Sanjie-Zhentong capsul-e.The difference of chemical compositions were Compared between in vivo and in vitr-o.By the research methods of serum pharmacochemistry,as normal rats for experimental animals,and prepare drug-containing plasma after rats were dosed by irrigating stomach.The UPLC/Q-TOF-MS were used to analyze and compare the differences of composition in Sanjie-Zhentong Capsule,the drug-containing plasma and blank plasma.And 20 com-pounds were detected in drug-containing plasma according to compare the compositions of the Sanjie-Zhentong capsule raw liquor,the plasma contained medicine and blank plas-ma.According to the retention time of the chromatographic peak conbining with the inf-ormations of the online spectrograms of UV,ESI-MS,ESI-MSMS,16 prototype compon-ents,1 possible metabolic components were identified and in addition 3 unknown compo-siteons would be necessary studied in the further research.The herbs' source and attribu-teion of the into-blood compositions were preliminary determined,binding to the analysi-s of single herb medicine ingredients in the compound of Sanjie-Zhentong Capsule in vit-ro.On the bases of this study,we used EMs rats uterus ectopic endometrial stromal cel-Is to research the effect of 8 into-blood compositions(Ginsenoside Rg1,Ginsenoside Rb1,Ginse-noside Rd,Notoginsenoside R1,7,4'-dihydroxyflavone,Loureinin B,Verticine,Verticinone)on the cells,which lay the foundation for the further study on the material basis of Sanjie-Zhentong capsule to the treatment of endometriosis.The results showed that,Ginse-noside Rb1?Ginsenoside Rd?Notoginsenoside R1?Loureinin B?7,4'-dihydrox-yflavone?Verticine?Verticin-one can inhibit the grovwth of endometrial cells;and Vertici-none can also inhibit the secretion of 6-keto-PGF1?TXB2,increase K/T ration,and can ease u-terine smooth muscle spasm,lessen the ?dysmenorrhea induced by end-ometriosis.Further tips,thesecomponents which were into blood(Ginsenoside Rb,,Ginsenoside Rd,Noto-ginsenoside R1,Loureinin B,7,4'-dihydroxyflavone?Verticine,Verti-cinone),may be the efficacy component of Sanjie-Zhentong capsule.
