Tripterygium wilfordii,which has anti-inflammatory,immunosuppressive and other pharmacological activities,and which has a significant effect,is widely used in the treatment of many autoimmune diseases such as rheumatoid arthritis,nephrotic syndrome,systemic lupus erythematosus,etc.and organ transplantation.Tripterygium wilfordii is toxic drugs,and its pharmacodynamic material basis is not entirely clear.Adverse reactions and therapeutic effect of different preparations on the market vary considerably.Thus,clinical application subject to certain restrictions.The extraction and purification technologies using the restricted organic solvents such as chloroform,methanol,and damage to the environment.In this paper,the method of fingerprint combing with pharmacodynamics,is uesd to study on ethanol extraction and macroporous resin purification process.The aim is to establish a extraction and purification technologies of Tripterygium wilfordii,which abandoned restricted organic solvents.In this paper,multi-target ingredients determination and fingerprint analysis were used as quality control method of extract and purification process.Studied on technology of jiangsumeitong,compared the fingerprints of the root,core root and root bark of Triptrygium wilfordiic,and then compared fingerprints of tripterygium preparations availarble on the market at last.We determined the root of Tripterygium wilfordii as materia medica of ours.Made an single-factor experiment to examine ethanol concentration,extraction time,extraction times,and other extraction conditions.And then MsPGN cell model was established to estimate different ethanol extactions.Orthogonal experimental design to optimize the extraction process,determined the extraction process were as follows:Tripterygium wilfordii were comminuted to coarse powder,95%the ratio of material to liquid was 1:10,95%ethanol solvent,reflux extract for 2 times,each time 1h.The validation tests showed that the average extraction rate of the index components was 74.10%.Studied on macroporous resin purification technologies,and established the initial purification technologies as follows:HPD 722 macroporous resin,ratio of diameter to height 1:10,resin material ratio 1:2,Resin quantity for mixing sample 1:10,wash the macroporous resin with 12 BV 20%ethanol at the rate of 12BV·h-1,and then eluted it with 12 BV 95%ethanol at the rate of 6BV·h-i.Rat adjuvant arthritis was established to evaluate the pharmacodynamics of 20%?40%?60%80%and 95%ethanol elutions.As to IL-1 content,all the elutions except 20%had significant deviation?p<0.05?compared to model group.As to degree of root swelling,60%?80%ethanol elutions had significant deviation?p<0.05?compared to model group.As to TNF-a content,60%ethanol elutions had significant deviation?p<0.05?compared to model group only.The result showed that pharmacodynamics effect of 60%elution was the best.Acute toxicity test indicated that LD50 of 60%elution was 382.58±77.19 mg·kg-1?...
