Objectives: Examine the role of AQP3 and Intestinal motility in the intervention of supplemented fluid-Increasing decotion for jin deficiency of slow transit constipation rat so as to explore the mechanism of laxative effect of supplemented fluid-Increasing decotion 。Methods:Sixty SD rats were randomly divided into six arms(n = ten in each group,half male and half female),including control arm(A),model arm(B),Itopride arm(C),supplemented fluid-Increasing decotion high dose arm(D),supplemented fluid-Increasing decotion medium dose arm(E)and supplemented fluid-Increasing decotion low dose arm 。 Afterwards,It was established for jin deficiency of slow transit constipation model by oral administration of compound diphenoxylate(1.5mg·kg-1·d-1)and water restriction(10ml·d-1)to rats in the B,C,D,E and F arms during seven days。Subsequently,To validate slow transit constipation model by comparing the first black stool discharge time,24 hours stool number and 24 hours fecal water content after ink oral administration。After that,suspension 4 ml of Itopride(50 mg·kg-1·d-1)was administered orally to rats in the C arms,which was replaced by equivalent volume supplemented fluid-Increasing decotion with the different dose(33.92、16.96、8.48g·kg-1·d-1)to rats in D,E and F arm,and the colons and blood were removed six days after same treatment 。Examine the expression of AQP3 as well as concentration of VIP and SP inthe proximal colon were detected by Immunohistochemistry and ELISA。Results:1、There was significant reduce in the aspect of 24 hours stool number and 24 hours fecal water content whereas the first black stool discharge time was appeared significantly decreased in model rats(P <0.01)。2、It was observed that Itopride and supplemented fluid-Increasing decotion(High,medium and low)could significantly decrease concentration of VIP,increase concentration of SP and down-regulate the expression levels of AQP3 in constipated rats(P<0.05)。 3 、 High dose of supplemented fluid-Increasing decotion in down-regulation of AQP3 protein expression level,decrease the concentration of VIP and increase concentration of SP was significantly better than the low dose,medium dose and itopride arms(P<0.05),but in arms between low dose,medium dose and itopride showed no significant difference(P>0.05)。Conclusion:1、By increasing the concentration of VIP,decreasing the concentration of SP to weaken colonic motility and Up-regulation of AQP3 protein expression was remarkable increased colonic absorption of water,it maybe the pathogenesis of jin deficiency slow transit constipation 。 2 、Supplemented fluid-Increasing decotion may leading to a laxative effect is:1)By decreasing VIP and increasing SP of concentration stimulates colonic contraction and peristalsis;2)By down regulation of AQP3 protein expression to inhibit water transport from the luminal to the vascular sdie 。 3 、Supplemented fluid-inereasing decotion in high-dose may be the optimal dose for clinical usage。...
